Las cepas de e. Coli productoras de toxina shiga se llaman stec. Se ha observado una alta incidencia de cepas setec aisladas del medio ambiente o de muestras cl\u00ednicas que son portadoras de m\u00e1s de un fago-stx. Por ello el principal objetivo de la tesis fue el estudio de la doble lisogenia doble en condiciones de laboratorio para poder entender el porqu\u00e9 de esta incidencia. Para ello se caracterizaron los fagos-stx aislados de diferentes cepas stec. Se realizaron estudios de lisogenia simple de estos fagos en cepas de e.Coli de laboratorio. Se obtuvieron fagos recombinantes que incorporaron un gen de resistencia a antibi\u00f3tico en medio del oper\u00f3n de la toxina shiga para marcarlos, de manera que se pudiera diferenciar entre el primer profago-stx y el segundo. Se realizaron los ensayos de lisogenia doble en los que se observ\u00f3 que la lisogenia doble ocurre m\u00e1s frecuentemente que la simple en condiciones de laboratorio, lo que explica esta alta incidencia de cepas stec portadoras de m\u00e1s de un fago-stx. Posteriormente se analiz\u00f3 el efecto de la doble lisogenia en la expresi\u00f3n y s\u00edntesis de la toxina shiga y en la liberaci\u00f3n de los fagos-stx. Se observ\u00f3 que la doble lisogenia mantiene el estado lisog\u00e9nico por una menor producci\u00f3n de fagos y de toxina. Posteriormente se analizaron los loci de inserci\u00f3n ocupados por los fagos-stx del estudio en diferentes hu\u00e9spedes. Los estudios confirman que el locus de inserci\u00f3n utilizado depende de la disponibilidad del mismo en la cepa en cuesti\u00f3n.
