La patog\u00e9nesis de a. Hydrophila es multifactorial. Con objeto de identificar genes implicados en la virulencia de la cepa ppd134\/91 de a. Hydrophila, se realizaron experimentos de sustracci\u00f3n g\u00e9nica, que llevaron a la detecci\u00f3n de 22 fragmentos de adn que codificaban 19 putativos factores de virulencia, incluyendo un gen que codificaba una prote\u00edna de sistema de secreci\u00f3n de tipo iii (t3ss). La creciente importancia del t3ss en la patog\u00e9nesis de diversas bacterias, llev\u00f3 a identificar y analizar la agrupaci\u00f3n g\u00e9nica del t3ss de las cepas h-1 y ah-3 de a. Hydrophila, as\u00ed como tambi\u00e9n el efector aext en la cepa ah-3. La inactivaci\u00f3n de los genes de t3ss aopb y aopd en a. Hydrophila ah-1, y asco en a. Hydrophila ah-3 conlleva una disminuci\u00f3n de la citotoxicidad, un incremento de la fagocitosis, y una reducci\u00f3n de la virulencia en diferentes modelos animales, demostrando estos resultados que el t3ss es necesario para patogenidad del g\u00e9nero aeromonas. El mutante en el gen aext de la capa ah-3 de a. Hydrophila muestra una ligera reducci\u00f3n de la virulencia, ensayada con diferentes m\u00e9todos. Por otro lado, la prote\u00edna aext present\u00f3 actividad adp-ribosiltransfereasa, y se demostr\u00f3 su translocaci\u00f3n v\u00eda t3ss al interior de c\u00e9lulas eucariotas, sistema que a su vez parece ser inducible in vitro en condiciones de depleci\u00f3n de calcio. Mediante el uso de diferentes sondas de adn, se determin\u00f3 la presencia del t3ss en cepas tanto cl\u00ednicas como ambientales de diferentes especies del g\u00e9nero aeromonas: a. Hydrophila, a. Varonil, y a. Caviae, y la codistribuci\u00f3n de esta agrupaci\u00f3n g\u00e9nica y del gen aext. Finalmente, con el fin de analizar la activaci\u00f3n transcricpcional de la agrupaci\u00f3n g\u00e9nica de t3ss y del efector aext en a. Hydrophila ah-3, los promotores predichos del oper\u00f3n aopn-aopd y el gen aext se fusionaron con el gen reporter gfp (green fluorescente prote\u00edn).
