El stress oxidativo, generado por el exceso patol\u00f3gico de especies reactivas de ox\u00edgeno, tambi\u00e9n parece participar en estos procesos. Esta tesis doctoral describe la conexi\u00f3n de ambos eventos en el proceso neurodegenerativo a trav\u00e9s de la ruta de se\u00f1alizaci\u00f3n akt \/ foxo3. En neuronas el stress oxidativo impide la acci\u00f3n neuroprotectora de igf-i bloqueando la fosforilaci\u00f3n inhibitoria del factor de transcripci\u00f3n foxo3 por la quinasa akt inducida por igf-i, en un proceso dependiente de la quinasa p38. Este bloqueo es necesario para permitir la activaci\u00f3n posterior de foxo3 por el stress oxidativo (v\u00eda jnks) y la muerte neuronal. Esta activaci\u00f3n de foxo3 contribuye a su vez a impedir su fosforilaci\u00f3n por akt inducida por igf-i. Este mecanismo pone de manifiesto por tanto dos hechos importantes: 1) la contribuci\u00f3n del stress oxidativo a la alteraci\u00f3n de la funci\u00f3n de factores neuroprotectores como igf-i y 2) la importancia del bloqueo de la se\u00f1alizaci\u00f3n de igf-i en el progreso de la neurodegeneraci\u00f3n inducida por stress oxidativo. Los astrocitos, c\u00e9lulas gliales de soporte neuronal, parecen presentar un mecanismo de defensa espec\u00edfico. En ellos el stress oxidativo tambi\u00e9n genera la muerte astrocitaria, pero a diferencia de neuronas, \u00e9ste no es capaz de bloquear la se\u00f1alizaci\u00f3n inhibitoria de igf-i sobre foxo3 (v\u00eda akt) sino que contrariamente la potencia, lo que favorece por tanto la supervivencia astrocitaria.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la tesis doctoral
