La 5-ht ejerce sus efectos a trav\u00e9s de su uni\u00f3n a receptores de membrana. La disponibilidad extracelular de 5-ht depende de un sistema de s\u00edntesis y liberaci\u00f3n (\u00abon\u00bb) y de un sistema de recaptaci\u00f3n (\u00aboff\u00bb). El transportador de serotonina (sert) se encarga de la captaci\u00f3n hacia el interior celular de la 5-ht y, en consecuencia, reduce la concentraci\u00f3n de 5-ht en el medio extracelular. Sert se ha convertido en una diana farmacol\u00f3gica efectiva en las patolog\u00edas en las que el sistema serotonin\u00e9rgico se encuentra alterado, tales como las alteraciones de la conducta y s\u00edndromes gastrointestinales. La mayor parte de los estudios realizados sobre sert se han realizado en modelos celulares heter\u00f3logos. Por ello, el primer objetivo del presente trabajo fue determinar si las c\u00e9lulas humanas de tipo enterocitario caco-2 expresaban end\u00f3genamente sert humano (hsert) y si pod\u00edan ser un modelo celular para su estudio. Los resultados muestran que las c\u00e9lulas caco-2 expresan hsert con caracter\u00edsticas funcionales, moleculares y de regulaci\u00f3n similares a las descritas para hsert de cerebro. Ello indica que esta l\u00ednea celular puede constituir un excelente modelo para el estudio de la regulaci\u00f3n fisiol\u00f3gica y farmacol\u00f3gica de hsert. A partir de este primer resultado se ha utilizado la l\u00ednea caco-2 para estudiar su regulaci\u00f3n fisiol\u00f3gica determinando el efecto del tratamiento a largo plazo con 5-ht sobre hsert. Los resultados muestran que la exposici\u00f3n de las c\u00e9lulas a 5-ht disminuye la funci\u00f3n de hsert debido a la interacci\u00f3n de 5-ht con el sitio alost\u00e9rico de baja afinidad de
