El pl\u00e1smidos aislado de la cepa b. Catenulatum l48 (pbc1) fue seleccionado para su posterior caracterizaci\u00f3n. El pl\u00e1smido se revel\u00f3 como una mol\u00e9cula circular de 2540 pares de bases con un contenido g+c del 64%. En el hipot\u00e9tico origen de replicaci\u00f3n se identific\u00f3 una repetici\u00f3n de 24 nucle\u00f3tidos repetida tres veces y media adem\u00e1s de cinco repeticiones invertidas, que recuerdan la organizaci\u00f3n de pl\u00e1smidos theta replicativos. El an\u00e1lisis de la secuencia tambi\u00e9n revel\u00f3 la presencia de tres orfs que codifican para p\u00e9ptidos mayores de 50 amino\u00e1cidos: repb, que codifica para la replicasa, un gen transcripcionalmente acoplado (orfx-like), y copg-like. Con el fin de estudiar las relaciones con pbc1, se realiz\u00f3 un \u00e1rbol filogen\u00e9tico en el que la prote\u00edna repb de pbc1 qued\u00f3 agrupada junto con las prote\u00ednas de replicaci\u00f3n de los pl\u00e1smidos pmb1 y pdojh10s, ambos procedentes de b. Longum, pero tambi\u00e9n con las prote\u00ednas rep de los pl\u00e1smidos cole2 y pxz10142) aislados de e. Coli y corynebacterium glutamicum, respectivamente, y en los que la replicaci\u00f3n tipo theta se ha demostrado experimentalmente. Experimentos de hibridaci\u00f3n mostraron que el replic\u00f3n de pbc1 no es com\u00fan entre las bifidobacterias. con el objeto de determinar el replic\u00f3n m\u00ednimo de pbc1 as\u00ed como la funcionalidad de las distintas orfs, diferentes fragmentos de pbc1 se amplificaron mediante pcr y se clonaron en el vector pbif, que por si s\u00f3lo no es capaz de replicar en bifidobacterias. Los fragmentos generados van desde el que engloba pr\u00e1cticamente toda la secuencia de pbc1 hasta el menor de todos que solamente incluye el gen repb y sus secuencias inmediatamente anteriores. Todos los derivados se mostraron capaces de replicar en bifidobacterias.
