Para estudiar los primeros estadios invasivos de esta neoplasia intratubular hemos desarrollado un modelo tumoral que se basa en los parecidos fenot\u00edpicos y funcionales de las c\u00e9lulas madre embrionarias (c\u00e9lulas es) con el cis. Para ello, marcamos una l\u00ednea e c\u00e9lulas es (ab1) transfect\u00e1ndola con el pl\u00e1smido de la prote\u00edna verde fluorescente (gfp). Posteriormente, trasplantamos dichas c\u00e9lulas al interior de los t\u00fabulos semin\u00edferos mediante microinyecci\u00f3n. Dejados a su evoluci\u00f3n estos trasplantes se transformaron en teratocarcinomas de aspecto histol\u00f3gico similar al de los tumores de las c\u00e9lulas germinales espont\u00e1neos. al ser la vascularizaci\u00f3n de gran importancia para la progresi\u00f3n tumoral, decidimos estudiar m\u00e1s en profundidad la arquitectura microvascular presente en los tumores experimentales generados, compar\u00e1ndola despu\u00e9s con la del test\u00edculo sano. Para ello, empleamos la t\u00e9cnica de los moldes de corrosi\u00f3n mediante perfusi\u00f3n de resinas acr\u00edlicas, por ser la que m\u00e1s detalles del sistema vascular revela a nivel morfol\u00f3gico. La densidad vascular de los tumores obtenidos result\u00f3 mayor que la del tejido testicular normal y los capilares neopl\u00e1sicos presentaron di\u00e1metros que doblaban los del \u00f3rgano. Igualmente, se estudi\u00f3 el origen celular del componente endotelial de los vasos neoformados, d\u00e1ndose la circunstancia de que era mixto, es decir, proced\u00edan tanto de las c\u00e9lulas trasplantadas como del hu\u00e9sped. asimismo, se estudi\u00f3 la reacci\u00f3n del estroma frente a la invasi\u00f3n tumoral. En las muestras histol\u00f3gicas de los tumores se evidenci\u00f3 una gran proliferaci\u00f3n de miofibroblastos, responsables de la secreci\u00f3n de col\u00e1genos, as\u00ed como la expresi\u00f3n de gran cantidad de factores relacionados con la vascularizaci\u00f3n, como son el vegf, pigf, vegf-r1, vegf-r2 y la metaloproteasa de la matriz mmp-2. por \u00faltimo, se analiz\u00f3 el efecto de la hipoxia (5%o2) sobre las c\u00e9lulas trasplantadas y las del propio tumor. En estos ensayos in vitro empleamos c\u00e9lulas madre de la l\u00ednea ab1 y las l\u00edneas f9 y p19, procedentes de teratocarcinomas experimentales.
