Sin embargo, s\u00f3lo las c\u00e9lulas fagociticas, c\u00e9lulas restrictivas para listeria monocytogenes disponen de ambos mecanismos mientras que las c\u00e9lulas primitivas s\u00f3lo poseen mecanismos no oxidativos. Para su caracterizaci\u00f3n hemos seguido una hip\u00f3tesis basada en que la estrategia de este pat\u00f3geno consist\u00eda en acumular el regulador temprano rab5a y excluir prote\u00ednas lisosomales, tales como lamp1 prote\u00edna asociada a membrana y la catepsina-d, proteasa lisosomal soluble. Lo que indicaba que algunas prote\u00ednas lisosomales podr\u00edan jugar un papel fundamental en la degradaci\u00f3n del pat\u00f3geno. De todas las posibles prote\u00ednas lisosomales hemos comenzado nuestro etudio por las proteasas lisosomales y de \u00e9stas por la familia de las catepsinas. De todas las catepsinas conocidas s\u00f3lo la aspartil-proteasa, catepsina-d y la ciste\u00edn proteasa se localizan en organelas, tales como endosomas o fogosomas mientras que el resto se encuentran bien extracelularmente o en el citosol. S\u00f3lo la catepsina-d pose\u00eda actividad listericida tanto en ausencia de c\u00e9lulas como en c\u00e9lulas permisivas de tipo fibroblasto, as\u00ed como en c\u00e9lulas restrictivas como son los macr\u00f3fagos de m\u00e9dula \u00f3sea. adem\u00e1s observamos una alta susceptibilidad en los ratones deficientes en catepsina-d a una infecci\u00f3n con este pat\u00f3geno comparado con ratones de fenotipo salvaje, lo que indicaba que dicha aspartil.Proteasa jugaba un papel fundamental en la respuesta inmune innata frente a dicho pt\u00f3geno. la capacidad listericida de la catepsina de la catepsina-d la localizamos a nivel fogosomal, la cual se dirig\u00eda contra el principal factor de virulencia de listeria monocytogenes: la listerolisina o o llo. los mon\u00f3meros de llo  se insertan en la membrana fagosomal produci\u00c2\u00bfndose una oligomerizaci\u00f3n de \u00e9stos y formando poros.
