Pinaster. Para poder estudiar su genoma es necesario disponer de forma f\u00edsica de su adn construyendo una genoteca gen\u00f3mica completa. Realizar esta tarea cuando se trabaja con genomas grandes y complejos es muy complicado ya que se requiere un gran n\u00famero de clones para disponer de una genoteca representativa. Para facilitar esta trabajo se ha optado por la utilizaci\u00f3n de los veh\u00edculos artificiales de bacterias (bac) ya que permiten la clonaci\u00f3n de insertos de gran tama\u00f1o. Hemos optimizado un m\u00e9todo para la construcci\u00f3n de una genoteca bac de p. Pinaster en el vector pindigo bac536, la genoteca parcial est\u00e1 formada por 72.192 clones con un tama\u00f1o medio de inserto de 107 kb. Hemos demostrado que la genoteca construida es v\u00e1lida para el rastreo con genes de inter\u00e9s para el investigador. De la misma forma, hemos construido una genoteca bac de p. Pinaster organizada en grupos de c\u00e9lulas que ocupa un espacio f\u00edsico muy inferior. Esta genoteca est\u00e1 formada por 83 grupos de c\u00e9lulas que representan 332.000 clones con un tama\u00f1o medio de inserto de 107 kb, hemos validado la genoteca en grupos de c\u00e9lulas con genes de inter\u00e9s. En el rastreo de ambas genotecas se han identificado una serie de clones bac con secuencia codificante para la fd-gogat y para una quitinasa de clase i. En resumen, hemos optimizado un m\u00e9todo para el estudio de genomas grandes y complejos como el que presenta el genoma del pino.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la tesis doctoral
