Para llevar a cabo este an\u00e1lisis se han probado distintos sistemas encaminados a la producci\u00f3n de diversas prote\u00ednas quim\u00e9ricas basadas en los ectodominios de e1 y e2, tanto completos como truncados. As\u00ed, utilizando el sistema basado en baculovirus se logr\u00f3 la expresi\u00f3n de la prote\u00edna e1340e2661, as\u00ed como la construcci\u00f3n e2661e1340, prote\u00edna quim\u00e9rica con los ectodominios permitados, que se obtuvo con un rendimiento m\u00e1s elevado. La caracterizaci\u00f3n espectrosc\u00f3pica (dicro\u00edsmo circular y fluorescencia), molecular (glicosilaciones y estado de  oligomerizaci\u00f3n) y antig\u00e9nica, revel\u00f3 que ambas prote\u00ednas pose\u00edan una estructura muy similar, encontr\u00e1ndose plegadas en una conformaci\u00f3n que se puede definir como nativa. a continuaci\u00f3n, se han determinado las propiedades fusog\u00e9nicas de e1340e2661 y e2661e1340, observ\u00e1ndose que ambas prote\u00ednas quim\u00e9ricas tienen la capacidad de desestabilizar ves\u00edculas constitu\u00eddas por fosfol\u00edpidos \u00e1cidos a ph 5, mientras que a ph 7 el efecto observado fue m\u00ednimo. Asimismo, mediante estudios con distintas l\u00edneas celulares se comprob\u00f3 que e\u00bb661e1340 interaccionaba con cd81 y sr-bi, prote\u00ednas propuestas como posibles receptores del hcv. Adicionalmente, utilizando drogas inhibidoras de diferentes rutas endoc\u00edticas se observ\u00f3 que la entrada de la prote\u00edna quim\u00e9rica en c\u00e9lulas de origen hep\u00e1tico puede tener lugar mediante diversos mecanismos como fagocitosis, macropinocitosis o mediante ves\u00edculas revestidas de clatrina.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la tesis doctoral
