Esta ruta consiste en un m\u00f3dulo de tres quinasas que consta de la mapk slt2, las mapkks mkk1 y mkk2 y la mapkkk bck1, que es activada por la prote\u00edna quinasa c, pkc1, y \u00e9sta a su vez por la gtpasa de peque\u00f1o tama\u00f1o, rho1. La activaci\u00f3n de esta ruta da lugar a la expresi\u00f3n de genes necesarios para el mantenimiento de la homeostasis de la pared celular mediada por el factor transcripcional rlm1. Esta ruta es la \u00fanica ruta de mapks en s. Cerevisiae que presenta dos mapkks, las cuales de hab\u00edan propuesto en estudios anteriores como redundantes. En este trabajo hemos comprobado que tanto mkk1 y mkk2, como prote\u00ednas quim\u00e9ricas obtenidas por el intercambio de sus dominios catal\u00edticos y regulatorios, son capaces de mantener eficientemente la transmisi\u00f3n de la se\u00f1al, confirmando que ambas tienen una funci\u00f3n equivalente en la ruta. Adem\u00e1s, en respuesta a la activaci\u00f3n de esta ruta, mkk1 y mkk2 son fosforiladas de manera independiente tanto de sus activadores como de la actividad de la propia mapk sit2. Hemos demostrado que sit2 es capaz de fosforilar in vitro a mkk1 y mkk2 y que la serina 50 de mkk2 es una diana directa de esta fosforilaci\u00f3n. Sin embargo, mkk2 es fosforilado en otros residuos adicionales que, aunque diferentes de los residuos consenso de fosforilaci\u00f3n por mapk, (s\/t)-p,dependen de sit2. Se ha caracterizado un dominio d (docking) consenso de interacci\u00f3n con mapks localizado en el extremo n-terminal de mkk2, esencial para su interacci\u00f3n con sit2 y para la retrofosforilaci\u00f3n. Nuestros datos muestran que las mapkks de la ruta de integridad celular son sustratos de su mapk, sugiriendo la existencia de un complejo mecanismo de regulaci\u00f3n por retroalimentaci\u00f3n a este
