Trabajos bioqu\u00edmicos previos realizados en nuestro laboratorio mostraron que la regi\u00f3n polib\u00e1sica del dominio c2 de la pkc alpha parec\u00eda desempe\u00f1ar un papel clave en el mecanismo de localizaci\u00f3n de dicha prote\u00edna en ves\u00edculas modelo en presencia de ptdins(4,5)p2. Por ello, el principal inter\u00e9s al inicio de esta tesis fue el de caracterizar el papel de esta regi\u00f3n polib\u00e1sica en el mecanismo de localizaci\u00f3n de la pkc alpha \u00abin vivo\u00bb, en c\u00e9lulas intactas. para ello, en primer lugar se caracteriz\u00f3 el mecanismo molecular de este isoenzima en c\u00e9lulas pc12 diferenciadas con ngf. Se sabe que estas c\u00e9lulas son un buen modelo de c\u00e9lulas nerviosas ya que responden a la estimulaci\u00f3n con ngf diferenci\u00e1ndose a neuronas, y tambi\u00e9n responden al neurotransmisor atp. En nuestros estudios demostramos que la estimulaci\u00f3n de dichas c\u00e9lulas con atp extracelular, activa a los receptores p2x de la membrana plasm\u00e1tica y permite la entrada de un pulso de calcio desde el exterior al interior de la c\u00e9lula. Este calcio es el motor que dirige la traslocaci\u00f3n de la pkc alpha a la membrana plasm\u00e1tica. Hemos demostrado, mediante el uso de mutantes de la pkc alpha, de inhibidores de la fosfolipasa c y de sondas de dag y ptdins(4,5)p2, que en la membrana,  la pkc se ancla por diversos residuos de su regi\u00f3n polib\u00e1sica, probablemente por su interacci\u00f3n con ptdins(4,5)p2. Estos resultados se corroboraron no s\u00f3lo en este sistema celular sino tambi\u00e9n en las c\u00e9lulas rbl-2h3, donde la fosfolipasa c tiene una mayor actividad y genera mayores cantidades de dag, y as\u00ed el ptdins(4,5)p2 est\u00e1 reducido.  Mediante el uso de mutantes en la regi\u00f3n polib\u00e1sica, se demostr\u00f3 que esta regi\u00f3n tambi\u00e9n en este tipo celular es esencial en el mecanismo de localizaci\u00f3n de la pkc alpha
