Adem\u00e1s, pgp es responsable de la aparici\u00f3n de la resistencia a m\u00faltiples f\u00e1rmacos (mdr) en l\u00edneas celulares tumorales, mediante la extrusi\u00f3n activa del f\u00e1rmaco antineopl\u00e1sico desde el citoplasma al medio extracelular. En este trabajo, se investiga la existencia de prote\u00ednas citos\u00f3licas que se unan a pgp y modulen la actividad de transporte de \u00e9sta, como una nueva posible l\u00ednea de dianas terap\u00e9uticas frente al c\u00e1ncer. Mediante la t\u00e9cnica de doble-h\u00edbrido en levadura, se descubren dos prote\u00ednas de las gtpasas de bajo peso molecular: rab4 y rab14, que interaccionan con pgp y podr\u00edan regular el transporte de pgp desde (o hacia) la membrana plasm\u00e1tica. Esta interacci\u00f3n se demuestra inicialmente in vitro. A continuaci\u00f3n, el trabajo demuestra en la l\u00ednea tumoral k562 y en su subl\u00ednea resistente (k562adr) que se presentan niveles opuestos de pgp y rab4: en la l\u00ednea resistente incrementa pgp en membrana y disminuye rab4. En sistemas de sobre-expresi\u00f3n transitoria de rab4 en k562adr, el incremento de rab4 se traduce en una disminuci\u00f3n de pgp en la membrana y a la quimiosensibilizaci\u00f3n de esta l\u00ednea celular. Por el contrario, cuando se disminuyen los niveles normales de rab4 y rab14 por silenciamiento g\u00e9nico, se incrementa pgp en la membrana. En conjunto, la interacci\u00f3n de rab4 y rab14 con pgp se postula como una nueva estrategia para la intervencion del fenotipo mdr mediado por pgp.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de
