La adaptaci\u00f3n a distintos ph implica la regulaci\u00f3n  y expresi\u00f3n de aquellos genes que le permitan crecer de forma \u00f3ptima en las nuevas condiciones ambientales. en asper gillus nidulans la regulaci\u00f3n por ph est\u00e1 mediada por el factor de transcripci\u00f3n con dedos de zinc pacc. El producto primario de la traducci\u00f3n, pacc72, permanece inactivo a ph \u00e1cido. A ph neutro o alcalino se produce la activaci\u00f3n de la ruta pal (com puesta por el producto de los genes pala, palb, palc, palf,  palh y  pali) de transducci\u00f3n de se\u00f1al, causando la activaci\u00f3n proteol\u00edtica de pacc72. La activaci\u00f3n requiere dos sucesos de prote\u00f3lisis. El primer corte proteol\u00edtico, dependiente de ph, ti ene lugar en una regi\u00f3n conservada denominada \u00c2\u00bfsignalling protease box\u00c2\u00bf. Se escinden 180 residuos desde el extremo carboxilo terminal de la prote\u00edna dando lugar a la forma pacc53. Es posible que esta proteasa \u00c2\u00bfse\u00f1alizadora\u00c2\u00bf sea palb. A continuaci\u00f3n, en una reacci\u00f3n independiente de ph ambiental, la proteasa \u00c2\u00bfprocesativa\u00c2\u00bf convierte a pacc53 en pacc27. las 6 prote\u00ednas de la ruta pal se dividen actualmente en dos complejos separados espacialmente en la c\u00e9lula. Un complejo \u00c2\u00bfsuperior\u00c2\u00bf de se\u00f1alizaci\u00f3n  por ph de la membrana plasm\u00e1tica y otro complejo \u00c2\u00bfinferior\u00c2\u00bf de se\u00f1alizaci\u00f3n del endosoma prevacuolar. El complejo \u00c2\u00bfsuperior\u00c2\u00bf est\u00e1 formado por las prote\u00ednas palf, palh y pali. El complejo \u00c2\u00bfinferior\u00c2\u00bf est\u00e1 formado por las prote\u00ednas pala, palc y como se  demuestra en esta tesis palb. Pala interacciona con vsp32\/sfn7, un componente del complejo escrt-iii (endosomal sorting complex required for transport), y con pacc72. Palb es una ciste\u00edn-proteasa de la familia de las calpa\u00ednas. A diferencia de su or t\u00f3logo en levaduras rim13, palb presenta un dominio mit \u00c2\u00bfmicrotubule interacting and trafficking\u00c2\u00bf en su extremo amino terminal. El dominio mit tambi\u00e9n est\u00e1 presente en la aaa-atpasa vps4, componente perif\u00e9rico del escrt-iii. En saccharomyces cerevisi ae, vps4 interacciona a trav\u00e9s de su dominio mit con vps46\/did2 una de las prote\u00ednas de la familia snf7. En a. Nidulans palb interacciona con vps24, componente del complejo escrt-iii, a trav\u00e9s del dominio mit de palb y en motivo mim (mit interacting motiv) de vps24, dicha interacci\u00f3n es espec\u00edfica y directa.  La interacci\u00f3n de palb con vps24 es responsable, al menos en parte, de la localizaci\u00f3n subcelular de palb.
