Para ello, en primer lugar se generaron y caracterizaron anticuerpos monoclonales espec\u00edficos frente a distintas regiones de la prote\u00edna, que han senido como herramientas para los estudios posteriores. Con dichos anticuerpos, se estudi\u00f3 la regulaci\u00f3n de la prote\u00f3lisis parcial de pten por caspasa-3. Se concluy\u00f3 que dicha prote\u00f3lisis se favorece por la actividad fosfatasa de l\u00edpidos de pten, mientras que se ve dificultada por su actividad fosfatasa de prote\u00ednas. Tambi\u00e9n se comprob\u00f3 que los anticuerpos frente al extremo c-terminal de pten pueden ser utilizados para inhibir la uni\u00f3n de \u00e9ste a dominios pdz. Asimismo, se realiz\u00f3 un escrutinio de interacciones de pten con otras prote\u00ednas en una l\u00ednea celular de c\u00e1ncer de mama (mcf-7). Se estudi\u00f3 la regulaci\u00f3n de la localizaci\u00f3n subcelular de pten por otro supresor tumoral, el inhibidor de cdks p27. Formas de pten de localizaci\u00f3n nuclear son retenidas en el citosol por p27. Finalmente, se realiz\u00f3 un estudio de la ruta pi3k\/pten\/akt utilizando como organismo modelo la levadura saccharomyces cerevisiae. Nuestros resultados indican que el fenotipo obsenado en la levadura depende de las subunidades catal\u00edticas y reguladoras de pi3k expresadas, de la actividad catal\u00edtica de los mutantes de pten expresados (cat\u00e1lisis intr\u00ednseca y cat\u00e1lisis in vivo) y de su nivel de expresi\u00f3n. Los resultados obtenidos en el sistema de la levadura reflejan lo que podr\u00eda ocurrir en c\u00e9lulas de mam\u00edfero. Debido a la simplicidad del sistema, \u00e9ste podr\u00eda ser empleado para estudios de alteraciones en varios punto de la ruta pi3k\/pten\/akt de mam\u00edferos, y para estudios funcionales de alto
