Aunque se desconocen las causas exactas de la degeneraci\u00f3n, se han propuesto diferentes mecanismos implicados como la excitotoxicidad, la p\u00e9rdida de soporte tr\u00f3fico y la neuroinflamaci\u00f3n. En esta tesis se han investigado los efectos de estos tres mecanismos en la viabilidad de las motoneuronas espinales. Tanto la excitotoxicidad por administraci\u00f3n de un inhibidor de la recaptaci\u00f3n de glutamato como la privaci\u00f3n de suero provocan la p\u00e9rdida de motoneuronas espinales en cultivo organot\u00edpico de m\u00e9dula espinal. Adem\u00e1s, el vegf protege de esta degeneraci\u00f3n via la pi3-k\/akt. Por otra parte, hemos demostrado que la citoquina proinflamatoria tnf-a potencia la excitotoxicidad glutamat\u00e9rgica de las motoneuronas espinales mediante, entre otros mecanismos, la inhibici\u00f3n del transportador de glutamato glt-1.
