Sin embargo, poco se conoce sobre las v\u00edas de se\u00f1alizaci\u00f3n ni sobre las causas que subyacen a la variabilidad de la respuesta entre pacientes. el objetivo general de esta tesis fue caracterizar las principales rutas de se\u00f1alizaci\u00f3n a trav\u00e9s de las cuales el bexaroteno ejerce su efecto antineopl\u00e1sico e identificar biomarcadores que permitan evaluar la susceptibilidad de los pacientes frente al tratamiento. Para ello, en nuestro planteamiento experimental decidimos caracterizar el efecto del bexaroteno en diferentes l\u00edneas celulares as\u00ed como de linfocitos de sangre perif\u00e9rica de pacientes con lcct para, posteriormente, ahondar en sus mecanismos moleculares de acci\u00f3n. nuestros resultados indican que el bexaroteno inhibe muy significativamente la capacidad clonog\u00e9nica de las c\u00e9lulas de lcct. Esta inhibici\u00f3n es el reflejo de su capacidad para inducir apoptosis pero, sobre todo, de su funci\u00f3n antiproliferativa, asociada  a la parada del  ciclo celular en los puntos  de control g1\/s y g2\/m. La acci\u00f3n del bexaroteno se encuentra regulada a dos niveles: inhibiendo la se\u00f1alizaci\u00f3n del factor nf-kappa b y mediante la activaci\u00f3n de p53\/p73, posiblemente a trav\u00e9s de atm. En este sentido hemos demostrado que la activaci\u00f3n de nf-kappa b en c\u00e9lulas de lcct se relaciona con la sensibilidad al bexaroteno; del mismo modo, la presencia de mutaciones en p53 o genes
