En cerebro de suicidas con depresi\u00f3n mayor, la densidad de los adrenoceptores alfa2a y alfa2c est\u00e1 aumentada. En cerebro de suicidas con depresi\u00f3n mayor, el tratamiento antidepresivo disminuye la densidad total del adrenoceptor alfa2a pero la fracci\u00f3n del adrenoceptor alfa2a en estado de alta afinidad permanece aumentada, contribuyendo, posiblemente, a la falta de respuesta al tratamiento antidepresivo. Por el contrario, el tratamiento antidepresivo regula a la baja el estado de alta afinidad del adrenoceptor alfa2c, pero no la densidad total del adrenoceptor alfa2c. La densidad del adrenoceptor \u00edY1 est\u00e1 aumentada en cerebro de suicidas con depresi\u00f3n mayor. El tratamiento antidepresivo disminuye la densidad del adrenoceptor \u00edY1 en suicidas con depresi\u00f3n mayor pero no en sujetos no deprimidos o en ratas. La supersensibilidad de los adrenoceptores alfa2a y alfa2c en depresi\u00f3n mayor no est\u00e1 asociada con los polimorfismos n251k o del322-325, respectivamente, pero el polimorfismo alfa2cdel322-325-adrenoceptor est\u00e1 asociado con la adicci\u00f3n a opi\u00e1ceos. Este hallazgo sugiere que la perdida de funcionalidad del adrenoceptor alfa2c conferida por el polimorfismo podr\u00eda subyacer los mecanismos neurobiol\u00f3gicos de la adicci\u00f3n a opi\u00e1ceos. Las prote\u00ednas rgs4 y rgs10 (regulator of g protein signalling), y las prote\u00ednas 14-3-3\u00edY y 14-3-3\u00c2\u00bf no est\u00e1n alteradas en cerebro de suicidas con depresi\u00f3n mayor, descartando su posible papel en la supersensibilidad del adrenoceptor alfa2. Sin embargo, la adicci\u00f3n a opi\u00e1ceos modula los niveles de las prote\u00ednas rgs4 y rgs10. La prote\u00edna rgs4 aparece aumentada tras la adicci\u00f3n a opi\u00e1ceos a largo plazo. Por el contrario, los niveles de la prote\u00edna rgs10 est\u00e1n disminuidos en corteza prefrontal de adictos a opi\u00e1ceos a corto plazo pero en los adictos a largo plazo los niveles vuelven al nivel control. Los niveles de las prote\u00ednas 14-3-3\u00edY y 14-3-3\u00c2\u00bf est\u00e1n aumentados en cerebro de suicidas con esquizofrenia. El tratamiento antipsic\u00f3tico normaliza los niveles de las prote\u00ednas 14-3-3\u00edY y 14-3-3\u00c2\u00bf y aumenta los niveles de la prote\u00edna rgs4.
