Est\u00e1 formado por seis subunidades (scorc1-6p) que se unen al dna, dirigiendo el correcto ensa mblaje de la maquinaria replicativa. Nuestro trabajo previo demostr\u00f3 que la subunidad scorc4p interacciona con chaperonas de la familia hsp70 o con su hom\u00f3logo dnak en escherichia coli. Mediante un an\u00e1lisis prote\u00f3mico del complejo formado entre scorc 4p y dnak, hemos encontrado en la segunda h\u00e9lice-? Del initiator specific motif (ism) de scorc4p una secuencia hidrof\u00f3bica (il4, residuos 184-188) que es la diana principal de estas chaperonas. La coexpresi\u00f3n en e. Coli de orc4p junto a orc1,2,3,5p y  cdc6p muestra que orc4p interacciona con orc1p, orc2p y orc5p y m\u00e1s d\u00e9bilmente con cdc6p. Realizamos mutag\u00e9nesis dirigida en el motivo il4 por cinco alaninas y hemos observado que si bien la mutaci\u00f3n no altera la estructura de orc4p, s\u00ed que afecta a  su interacci\u00f3n con la subunidad orc2p. Por otro lado, la sustituci\u00f3n al\u00e9lica en orc4 de mutantes en cada residuo del motivo il4 muestra que mientras que la mutaci\u00f3n l184a es letal para la c\u00e9lula, las mutaciones l185a y l186a dan como resultado un fe notipo termosensible que hace que estas estirpes tengan un defecto en el inicio de replicaci\u00f3n. En resumen, estos resultados nos hacen postular que las chaperonas de la familia hsp70 desempe\u00f1an un papel importante en el ensamblaje del complejo orc.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos
