Estas se\u00f1ales pueden venir de mol\u00e9culas solubles, de mol\u00e9culas que conforman la matriz extracelular o a trav\u00e9s del contac to directo con otras c\u00e9lulas, y son recibidas por receptores espec\u00edficos celulares. La activaci\u00f3n de estos receptores inicia multitud de rutas de se\u00f1alizaci\u00f3n. Se ha realizado un gran esfuerzo en el estudio de estas rutas para ahondar en el conocimie nto de su funci\u00f3n celular y porque su mal funcionamiento est\u00e1 ligado a numerosas enfermedades. Gracias a estos trabajos cada vez se tiene m\u00e1s claro que la transducci\u00f3n de la se\u00f1al no ocurre de manera lineal, desde los receptores hasta sus respuestas celulares espec\u00edficas, sino que existe una vasta interconexi\u00f3n entre distintas rutas de se\u00f1alizaci\u00f3n. Muchas de las mol\u00e9culas que forman parte de esta densa red son prote\u00ednas modulares que poseen una regulaci\u00f3n compleja. En el desarrollo de esta tesi s se ha estudiado la estructura y regulaci\u00f3n mediante microscop\u00eda electr\u00f3nica de tres de estas prote\u00ednas, involucradas en las rutas de se\u00f1alizaci\u00f3n de la superfamilia ras. Se trata de:  \u00c2\u00bf\tvav3- factor intercambiador de nucle\u00f3tidos (gef, guanine nucle otide exchage factor) que activa a prote\u00ednas pertenecientes a la familia de gtpasas (gtp hidrolasas) rho.  \u00c2\u00bf\tsyk (spleen tyrosine kinase)- prote\u00edna tiros\u00edn quinasa responsable, entre otras cosas de la fosforilaci\u00f3n y activaci\u00f3n de vav.   \u00c2\u00bf\tepac2 (exc hange protein directly activated by cyclic amp)- es un gef, al igual que vav3, que intercambia nucle\u00f3tidos de guanina a prote\u00ednas de la familia de gtpasas
