eficaz frente a plasmodium falciparum y la capaciedad de par\u00e1sito de desarrollar resistencia frente a los f\u00e1rmacos antimal\u00e1rios sugieren , como estrategia en la investigaci\u00f3n, la exploraci\u00f3n de nuevas dianas funcionales que permiten el desarrollo de nuevos abordajes terap\u00e9rutocos frenta ala malaria.La validaci\u00f3n de un gen , \u00f3 de su producto , como diana requere conocer la funci\u00f3n del mismo dentro de la biolog\u00eda del par\u00e1sito.En este sentido, se han venido desarrollando distintos sistemas para el an\u00e1lisis funcionales de genes de p.Falciparum como la inactivaci\u00f3n de genes por recombinaci\u00f3n , los sistemas de silenciamiento mediante rna antisentido \u00f3 mediante rna interferente.Los objetivo de este trabajo de investigaci\u00f3n han sido:la selecci\u00f3n de genes modelo en p.Falciparum que pudiesen representar dianas espec\u00edficas par el dise\u00f1o de nuevos agentes antimal\u00e1ricos , explorar distintos sistemaas de silenciamiento de genes en p.Falciparum, para lo que ha sido necesario la construcci\u00f3n de una colecci\u00f3n de vextores, el dise\u00f1o de oligoribonucle\u00f3tidos cortos y la optimizqaci\u00f3n de sistemas de transfecci\u00f3n , y pro \u00faltimo desarrollar sistemas molecualres de an\u00e1lisis del efecrto funcional del silenciamiento en el ciclo biol\u00f3gico del par\u00e1sito.  la introducci\u00f3n y expresi\u00f3n de material gen\u00e9tico ex\u00f3geno en plasmodium falciparum es transitoriamente eficiente, pero la baja segragaci\u00f3n de pl\u00e1smidos supone un grave impedimento par la expresi\u00f3n a largo plazo y el establecimiento de clones del par\u00e1sito.El silenciamiento mediante interrupci\u00f3n por recombinaci\u00f3n hom\u00f3loga para an\u00e1lisis de la funci\u00f3n de genes cuanta con importantes inconvenientes experimentales.Es posible el silenciamiento parcial de genes mediante interferenca con rna, bien como rna antisentido o como rna de doble cadena de paque\u00f1o tama\u00f1o, que parecen inhibir su traducci\u00f3n por un mecanismo compartido ,el silenciamiento<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la tesis
