Asimismo se estudia el mecanismo de toxicidad de la azatioprina.  en la primera parte de esta tesis se muestra un m\u00e9todo para la determinaci\u00f3n de la actividad tpmt y el estudio de la actividad del enzima en la poblaci\u00f3n espa\u00f1ola. Este resultado es de una gran relevancia a la hora de mejorar la seguridad y eficacia del tratamiento cl\u00ednico con azatioprina, pues se incluye una gu\u00eda para la dosificaci\u00f3n del f\u00e1rmaco en funci\u00f3n de la actividad individual de esta enzima.  en segundo lugar, se estudia el mecanismo de toxicidad de la azatioprina en un modelo de hepatocitos de rata en cultivo primario, encontr\u00e1ndose que el f\u00e1rmaco induce necrosis por un mecanismo mediado por la permeabilidad mitocondrial transitoria y activaci\u00f3n jnk y p38 quinasa. Se encuentra que es la depleci\u00f3n de glutation la responsable de la toxicidad y que el cotratamiento azatioprina\/n-acetilciste\u00edna mejora la metabolizaci\u00f3n del f\u00e1rmaco y previene completamente la toxicidad de \u00e9ste en nuestro modelo de hepatocitos en cultivo.  este \u00faltimo hallazgo puede tener importantes repercusiones cl\u00ednicas, ya que la dosificaci\u00f3n
