Los pacientes no estuvieron bajo ning\u00fan tipo de tratamiento conantibi\u00f3ticos en el momento de la cirug\u00eda abdominal y la presencia de neoplasia fue motivo de exclusi\u00f3n. El estudio fue aprobado por el comit\u00e9 \u00e9tico del hospital y se solicit\u00f3 el consentimiento de cada paciente participante en el estudio. El grupo de donantes de tejido comprendi\u00f3 18 pacientes. Tras la intervenci\u00f3n quir\u00fargica el tejido adiposo fue transportado inmediatamente al laboratorio bajo condiciones est\u00e9riles, en una soluci\u00f3n krebs-ringer-hepes (krh) con la siguientes composici\u00f3n en mm\/i: nacl, 125; kci, 5; mgso4, 1.2; cacl2 2; kh2po4, 1.2; glucosa, 6; hepes, 25 (ph 7.4, ajustado con naoh). Una vez el tejido en el laboratorio, se eliminaron vasos sangu\u00edneos y tejido conectivo, se lav\u00f3 con krh, se cort\u00f3 en fragmentos (300 a 400 mg de tejido por pocillo) y se realizaron varios lavados m\u00e1s con krh hasta que el tejido estuvo totalmente limpio. Los fragmentos de tejido se colocaron en placas de 6 pocillos a las que se a\u00f1adieron 2.5 ml de medio dmem suplementado con bicarbonato s\u00f3dico (3.7 g\/l), glutamina (2mm), piruvato s\u00f3dico (1mm),p enicilina (100 u\/ml), estreptomicina (100 ug\/ml) y suero fetal de ternera (0.5%). Tras un per\u00edodo de incubaci\u00f3n de 24 horas a 37\u00c2\u00ba c en un incubador de co2 con un autocalibrado peri\u00f3dico (heraeus), bajo una atm\u00f3sfera humidificada de 95% aire &#8211; 5%
