La degradaci\u00f3n intracelular de prote\u00ednas tiene implicaciones fisiol\u00f3gicas y patol\u00f3gicas importantes. Los dos sistemas proteol\u00edticos m\u00e1s importantes en c\u00e9lulas eucariotas, son los proteasomas y los lisosomas. De entre las v\u00edas de degradaci\u00f3n lisosomal de prote\u00ednas, la v\u00eda autof\u00e1gica mediada por carabinas es un proceso selectivo a\u00fan muy desconocido. El primer objetivo de esta tesis consiste en estudiar la conformaci\u00f3n de las prote\u00ednas citos\u00f3licas que atraviesan la membrana del lisosoma para su degradaci\u00f3n selectiva. Empleando dhfr, con o sin metotrexato sintetizada in vitro y marcada con 35s, y lisosomas, aislados de h\u00edgado de ratas en ayuno prolongado, determinamos que el metotrexato afecta la entrada de dhfr (que sigue una v\u00eda de degradaci\u00f3n lisosomal mediante autofagia mediada por carabinas) a los lisosomas, pero no su uni\u00f3n a la membrana del lisosoma. El segundo objetivo de esta tesis permiti\u00f3 el estudio de una secuencia similar a kferq (secuencia que se postula dirige la degradaci\u00f3n selectiva) en la prote\u00edna gapdh (excelente sustrato de la v\u00eda autof\u00e1gica mediada por carabinas), observando que en concreto el amino\u00e1cido glutamina de esta secuencia, y por tanto la secuencia completa, no es importante en la se\u00f1alizaci\u00f3n de esta prote\u00edna para su degradaci\u00f3n selectiva, mientras que la conformaci\u00f3n de la prote\u00edna parace crucial en su degradaci\u00f3n. En el tercer objetivo de esta tesis doctoral estudiamos la importancia de la prote\u00edna lamp2 como receptor de la v\u00eda autof\u00e1gica mediada por carabinas, observando que los datos obtenidos en fibroblastos embrionarios de ratones (control, deficientes en lamp1, deficientes en lam2, y deficientes en lamp1 y en lamp2) no apoyan el papel de esta prote\u00edna como receptor de la v\u00eda
