Las prote\u00ednas influyen en m\u00faltiples procesos fisiol\u00f3gicos, desde el ciclo celular y la apoptosis hasta la formaci\u00f3n del hueso y el establecimiento de rutas neuronales. adem\u00e1s, la actividad de las proteasas est\u00e1 involucrada en el desarrollo de diversas enfermedades, incluyendo la formaci\u00f3n de met\u00e1stasis tumorales. en la presente tesis doctoral se describe la identificaci\u00f3n y caracterizaci\u00f3n de nuevas proteasas, la matriptasa-2 y la poliserasa-1, pertenecen al grupo de las ser\u00edn-proteasas transmembrana de tipo ii. El gen de la matriptasa-2 se expresa espec\u00edficamente en h\u00edgado, mientras que el gen de la poliserasa-1 se expresa en m\u00faltiples tejidos y l\u00edneas celulares tumorales. La estructura de la poliserasa-1 se caracteriza por la presencia de dos dominios catal\u00edticos de tipo ser\u00edn-proteasa y un dominio hom\u00f3logo inactivo del mismo tipo. Los dominios catal\u00edticos recombinantes de la matriptasa-2 y la poliserasa-1 muestran actividad proteol\u00edtica frente a sustratos t\u00edpicos de las ser\u00edn-proteasas. adem\u00e1s, se describe la identificaci\u00f3n y caracterizaci\u00f3n de 22 nuevas prote\u00ednas de la familia usp de ciste\u00edn-proteasas espec\u00edficas de ubiquitina, incluyendo cuatro hom\u00f3logos inactivos. Los genes de estas nuevas proteasas se expresan preferentemente en m\u00fasculo esquel\u00e9tico y test\u00edculo, aunque algunas usps se expresan en m\u00faltiples tejidos. Las formas recombinantes de doce de estas nuevas usps son activas proteol\u00edticamente frente a un sustrato que contiene ubiquitina. Ninguno de los hom\u00f3logos inactivos muestra actividad proteol\u00edtica en este ensayo.
