{"id":70406,"date":"2018-03-09T23:14:55","date_gmt":"2018-03-09T23:14:55","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/localizacion-subcelular-y-regulacion-post-traduccional-del-transportador-hepatico-de-adenosina-rcnt2\/"},"modified":"2018-03-09T23:14:55","modified_gmt":"2018-03-09T23:14:55","slug":"localizacion-subcelular-y-regulacion-post-traduccional-del-transportador-hepatico-de-adenosina-rcnt2","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/bioquimica-molecular\/localizacion-subcelular-y-regulacion-post-traduccional-del-transportador-hepatico-de-adenosina-rcnt2\/","title":{"rendered":"Localizaci\u00f3n subcelular y regulaci\u00f3n post-traduccional del transportador hep\u00e1tico de adenosina rcnt2."},"content":{"rendered":"<h2>Tesis doctoral de <strong> Sylvie Duflot <\/strong><\/h2>\n<p>Las funciones del h\u00edgado son importaciones debido a su localizaci\u00f3n estrat\u00e9gica, entre el sistema digestivos y el resto del cuerpo (la circulaci\u00f3n sist\u00e9mica).La generaci\u00f3n de los anticuerpos contra los transportadores de nucle\u00f3sidos sodio-dependientes rcnt1 (pirimidina preferente)y rcnt2 (purina preferente) permitieron identificar la presencia de estos dos trasportadores en h\u00edgado.El primer objetivo de esta tesis es establecer la localizaci\u00f3n y la ditribuci\u00f3n subcelular de rcnt1-y rcnt2 en hepatocitos de rata.  los hepatocitos representan un 60-80% de la masa celular totaldel h\u00edgado:son c\u00e9lulas altamente polarizadas y se caracterizan po tener una membrana plasm\u00e1tica diferenciada en tres dominios funcionales diferentes:la membrana basal la apical y la lateral.Por estas caracter\u00edsticas y po ser muy activos metabolitamente,las c\u00e9lulas hep\u00e1ticas son un buen modelo para el estudio de las rutas endoc\u00edticas.Examinamos la localizaci\u00f3n sbcelular de tres maneras distintas.Primero , detectmos por t\u00e9cnica de transporte y por western blot la expresi\u00f3n de cnt1 y cnt2 en fraciones purificadas de endosomas de h\u00edgado de rata ,mediante utilizaci\u00f3n de anticuerpos contra los transtornos . Tercio por t\u00e9cnicas de microscop\u00eda electr\u00f3nica .Estos estudios indican que el transportador de nucleosidos cnt1 llega en la membrana canalicular, en la cual el transportador es biol\u00f3gicamente activo, por una ruta indirecta a trav\u00e9s de la membrana basolateral. Su inserci\u00f3n se realiza cerca de microdominios especilizados enriquecidos en caveolas. Cnt2 se detecta de manera casi exclusiva en preparaciones enriquecidas en membranas plasm\u00e1ricas basolaterales en las cuales el transportador es activo.Estos estudios nos indican que los dos transportes cnt1 y cnt2 tienen una ubicaci\u00f3n diferente en c\u00e9lulas hep\u00e1ticas, por lo que estos dos transportadores parecen estar diferenicalmente regulados.En la segunda parte de esta
