{"id":70724,"date":"2004-08-10T00:00:00","date_gmt":"2004-08-10T00:00:00","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/estudio-de-los-mecanismos-de-mutagenesis-y-potencia-mutagenica-de-quinolonas\/"},"modified":"2004-08-10T00:00:00","modified_gmt":"2004-08-10T00:00:00","slug":"estudio-de-los-mecanismos-de-mutagenesis-y-potencia-mutagenica-de-quinolonas","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/ciencias-medicas\/estudio-de-los-mecanismos-de-mutagenesis-y-potencia-mutagenica-de-quinolonas\/","title":{"rendered":"Estudio de los mecanismos de mutag\u00e9nesis y potencia mutag\u00e9nica de quinolonas"},"content":{"rendered":"<h2>Tesis doctoral de <strong>  Gonz\u00e1lez Cabeza Jos\u00e9 Guillermo <\/strong><\/h2>\n<p>Dada la importancia cl\u00ednica de las quinolonas en el el control de enfermedades infecciosas y las posibles implicaciones que puede tener el hecho de que estas mol\u00e9culas sean agentes mutag\u00e9nicos en bacteriasl, el presente estudio se ha centrado en profundizar en los mecanismos de mutag\u00e9nesis, utilizando la ciprofloxacina como mol\u00e9cula modelo, y en determinar el potencial mutag\u00e9nico de quinolonas de actual uso cl\u00ednico.  en lo que se refiere a los mecanismos de mutag\u00e9nesis, se ha demostrado que para la mutag\u00e9nesis inducida por ciprofloxacina, se requiere que las bacterias posean un sistema de reparaci\u00f3n de escisi\u00f3n de nucle\u00f3tidos (ner) funcional, descart\u00e1ndose que el oper\u00f3n moa, el cula codifica genes de bios\u00edntesis de cofactores de molibdeno tenga alg\u00fan papel en dicha mutag\u00e9nesis.  estos resultados corroboran estudios anteriores, los cuales suger\u00edan la participaci\u00f3n del sistema ner en dicha mutag\u00e9nesis, e indican que probablemente otros genes deleccionados en las cepas de ensayo de s.Enterica typhimurium ta104 y ta2659 distintos del uvrb, no deben tener un papel importante en la mutag\u00e9nesis mediada por las quinolonas.  por otra parte, se ha demostrado que la cicprofloxacina puede generar alguna especie reactiva de ox\u00edgeno, probablemente el ani\u00f3n super\u00f3xido (o2-) y\/o ox\u00edgeno singlete produciendo da\u00f1o oxidativo, el cual no debe de ser preferentemente la lesi\u00f3n 8-oxog.  el conjunto de estos resultados indica que las quinolonas deben de producir diferentes tipos de lesiones en el dna de las c\u00e9lulas bacterianas. As\u00ed, la interacci\u00f3n de la mol\u00e9cula de quinolona con el complejo dna-dna girasa debe generar un tipo de distorisi\u00f3n an\u00e1loga a un enlace intercatenario, dando lugar a una lesi\u00f3n premutag\u00e9nica. Dicha lesi\u00f3n puede ser procesada por el sistema ner y convertirse en una lesi\u00f3n mutag\u00e9nica sobre la cual actuar\u00eda una dna polimerasa tendente al error, an\u00e1loga a mucab y que introducir\u00edra una mutaci\u00f3n.
