Carmen Duran Ruiz <\/strong><\/h2>\n<p>La aterosclerosis es una enfermedad cr\u00f3nica normalmente resultado de la respuesta inflamatoria del organismo en la que las c\u00e9lulas que forman parte de la pared arterial, c\u00e9lulas musculares lisas y c\u00e9lulas endoteliales en mayor medida, interact\u00faan con lipoprote\u00ednas y c\u00e9lulas inflamatorias (macr\u00f3fagos y linfocitos t) derivando en la formaci\u00f3n de la placa de atheroma. Debido al enrome potencial que el an\u00e1lisis prote\u00f3mico parece presentar, la electroforesis bidimensional (2de) y espectrometr\u00eda de masas (em) fueron seleccionadas como la \u00abmaquinaria\u00bb esencial para este an\u00e1lisis, con el objeto de entender qu\u00e9 ocurre, a nivel proteico, en el desarrollo de la enfermedad ateroscler\u00f3tica. en el transcurso de esta tesis se han considerado dos estrategias diferentes. en un principio se plante\u00f3 el estudio del proteoma de algunos de los componentes de la placa , entre ellos las c\u00e9lulas endoteliales, que fueron tratadas con agentes proinflamatorios (il-1beta) con objeto de caracterizar aquellas prote\u00ednas, expresadas en estas c\u00e9lulas sometidas a condiciones inflamatorias, que podr\u00edan estar directamente implicadas en la formaci\u00f3n posterior de la placa de atheroma. Posteriormente, la posibilidad de trabajar directamente con placa de ateroma extra\u00edda mediante endarterectom\u00eda carot\u00eddea, permiti\u00f3 estudiar, mediante an\u00e1lisis prote\u00f3mico, el secretoma d la placa de ateroma. mediante esta metodolog\u00eda exvivo, hemos demostrado, en primer lugar, que las placas de ateroma cultivadas in vitro son capaces de secretar prote\u00ednas al medio, pudiendo ser detectadas mediante 2de. La mayor\u00eda de estas prote\u00ednas eran secretadas en mayor intensidad en presencia de placa de ateroma en comparaci\u00f3n con los segmentos arteriales tomados como controles. Adem\u00e1s, los niveles de secreci\u00f3n eran mayores en aquellas regiones en los que se detectaba una mayor progresi\u00f3n de la patolog\u00eda.
