Vrk1 es una prote\u00edna nuclear que fosforila a diversos factores de transcripci\u00f3n in vitro  como el supresor de tumores p53. En el presente trabajo abordamos la caracterizaci\u00f3n funcional de vrk1, la relaci\u00f3n entre el supresor de tumores p53 y vrk1 e investigamos las funciones fisiol\u00f3gicas de vrk1 en la c\u00e9lula, as\u00ed como su posible papel en procesos tumorales. Vrk1 es una quinasa ampliamente expresada en distintas l\u00edneas celulares tanto tumorales como normales y durante todo el ciclo celular. Mediante experimentos de cotransfecci\u00f3n mostramos que vrk1 induce la acumulaci\u00f3n nuclear y actividad transcripcional de p53. Esta estabilizaci\u00f3n es debida a un efecto postraduccional sobre p53 y dependiente de la actividad quinasa de vrk1 aunque independiente de la acci\u00f3n de la quinasa de \u00abcheckpoint\u00bb chk2. Vrk1 fosforila in vitro a p53 en la treonina-18 dentro de la regi\u00f3n de uni\u00f3n a mdm2 y la \u00fanica fosforilaci\u00f3n de p53 por la quinasa interrumpe la interacci\u00f3n entre estas dos prote\u00ednas in vitro. Vrk1 nos fosforila a mdm2. In vivo el efecto de vrj1 sobre p53 es, al menos parcialmente, independiente de mdm2. La sobreexpresi\u00f3n de vrk1 promueve la fosforilaci\u00f3n en treonina-18 de p53 y su acetilaci\u00f3n por parte del coactivador p300, lo que podr\u00eda explicar el aumento de la actividad transcripcional. La supresi\u00f3n de la expresi\u00f3n de vrk1 en l\u00edneas humanas provoca un fenotipo de disminuci\u00f3n de la proliferaci\u00f3n. La prote\u00edna vrk1 es encontrada en diferentes niveles en tumores, y altos niveles de la quinasa correlacionan con altos niveles de p53 y genes de respuesta a p53 en algunos tumores, as\u00ed como con algunos marcadores t\u00edpicos de proliferaci\u00f3n. La familia vrk tambi\u00e9n est\u00e1 regulada durante el desarrollo hematopoy\u00e9tico murino.
