Sus implicaciones en biomedicina las ha convertido en dianas muy atractivas para estudios estructura-funci\u00f3n. La  -1,3-galactosiltransferasa, objeto de la presente tesis, es la responsable de la s\u00edntesis del disac\u00e1rido terminal gal 1,3gal-, el ant\u00edgeno mayoritario en el xenotransplante.  en la actualidad, la disponibilidad de \u00f3rganos humanos para transplantes supone un grave problema dada la creciente demanda de \u00f3rganos y la escasez de donantes. El xenotransplante, o transplante de \u00f3rganos animales a humanos, podr\u00eda ser una soluci\u00f3n transitoria a este problema. Sin embargo, el mayor inconveniente del xenotransplante es el rechazo hiperagudo causado por la respuesta del sistema inmunitario a ant\u00edgenos espec\u00edficos de los tejidos animales. El ant\u00edgeno mayoritario es el ep\u00edtopo  gal o xenoant\u00edgeno, que se expresa abundantemente en la superficie de las c\u00e9lulas endoteliales de todos los mam\u00edferos, excepto los humanos y los monos del viejo mundo. el ep\u00edtopo  gal es producido por el enzima  -1,3-galactosiltransferasa ( 3gt). Se han propuesto diferentes estrategias para evitar el rechazo hiperagudo: a) manipulaci\u00f3n gen\u00e9tica de los animales donantes (cerdos), para que produzcan \u00f3rganos que no expresen el ep\u00edtopo, b) neutralizaci\u00f3n de los anticuerpos naturales humanos con estructuras sint\u00e9ticas que contengan el ep\u00edtopo, c) eliminaci\u00f3n de los anticuerpos previo al transplante, d) bloqueo del substrato aceptor por sobreexpresi\u00f3n de otras glicosiltransferasas en el animal donante, y e) inhibici\u00f3n de la actividad enzim\u00e1tica  3gt en el tejido a transplantar.  el objetivo de este trabajo es la caracterizaci\u00f3n molecular de la  3gt bovina. La investigaci\u00f3n se centra en el estudio estructura-funci\u00f3n de su mecanismo catal\u00edtico, utilizando ingenier\u00eda de prote\u00ednas y t\u00e9cnicas de enzimolog\u00eda.
