La funci\u00f3n primordial de esta prote\u00edna es promover la adsorci\u00f3n de los fosfol\u00edpidos del surfactante (principalmente dppc) desde la hipofase acuosa a la interfase aire-l\u00edquido en el interior del alv\u00e9olo, donde produce una dr\u00e1stica disminuci\u00f3n de la tensi\u00f3n superficial evitando el colapso alveolar durante la espiraci\u00f3n y facilitando el trabajo pulmonar. La sp-b nativa, que posee 3 puentes disulfuro intracatenarios, se a\u00edsla como un homod\u00edmero de elevada hidrofobicidad, estabilizado por un puente disulfuro intercatenario. Partiendo de sp-b porcina nativa, se ha optimizado la producci\u00f3n de una forma reducida y carboxiamidometilada de la prote\u00edna, cuya caracterizaci\u00f3n ha permitido concluir que la sp-b as\u00ed modificada mantiene los determinantes estructurales esenciales para el desarrollo de su funci\u00f3n tensioactiva in vitro. Estos resultados fueron el punto de partida par ala producci\u00f3n de prote\u00edna en un organismo como e.Coli, con el doble objetivo de optimizar un sistema de producci\u00f3n de sp-b madura y de obtener el precursor de la prote\u00edna (prosp-b), que no puede aislarse de fuentes naturales. La estructura y propiedades de interacci\u00f3n l\u00edpido-prote\u00edna dela prosp-b recombinante aislada y purificada ha sido analizada mediante diversas t\u00e9cnicas biof\u00edsicas.  a lo largo de la v\u00eda exoc\u00edtica de los neumocitos, el precursor de sp-b sufre dos cortes proteol\u00edticos para eliminar dos prop\u00e9ptidos n- y c- terminales. en esta ruta se produce un progresivo descenso de ph cuyo efecto sobre el comportamiento de la prosp-b recombiante ha sido analizado a diversos niveles, con el empleo de otra variante del precursor de sp-b producida tambi\u00e9n en e.Coli, y de dos p\u00e9ptidos sint\u00e9ticos dise\u00f1ados a partir de la secuencia de cierta regi\u00f3n de la prote\u00edna.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la tesis doctoral
