Se ha observado que los inhibidores de la hmg-coa reductasa (estatinas) son utiles en la reversion de la disfuncion endotelial. tanto la via de la l- arginina -oxido nitrico -cgmp como de la endotelina estan involucradas en la regulacion del tono vascular. En este trabajo se muestra que las estatinas previenen la inhibicion de la expresion de la oxido nitrico sintasa endotelial inducida por ldl oxidadas. Asimismo, la expresion del gen de la pre-proendotelina-1 es inhibida por las estatinas por un mecanismo transcripcional. La interrupcion de la via biosintetica del mevalonato-colesterol, y la subsiguiente inhibicion de la geranilgeranilacion de determinadas proteinas, reduce los niveles de transcripcion de la pre-proendotelina-1. ademas, hemos encontrado que la actividad de las gtpasas rhoa y rhob, en cuya maduracion esta involucrada la geranilgeranilacion, es requerida para mantener la expresion basal del gen de la pre-proendoetlina-1. Por ultimo, hemos mostrado como la simvastina inhibe la actividad transcripcional del gen de la pre-proendotelina-1 mediante la afectacion de la via de rho. por ello, pensamos que el control de tono vascular y la respuesta proliferativa mediada por et-1 esta regulada a multiples niveles, entre los que la familia de proteinas rho  juega un papel esencial.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos
