Cataliza la descarboxilaci\u00f3n oxidativa del isocitrato en a-cetoglutarato en presencia de nad(p)+ y liberando co2 en el proceso. En el organismo hal\u00f3filo haloferax volcanii, esta enzima es dim\u00e9rica, dependiente de nadp+ y de la presencia de mg2+ como cati\u00f3n divalente, siendo estable en un amplio rango de concentraciones salinas. En la actualidad existen muchas isocitrato deshidrogenasas secuenciadas pero muy pocas estructuras tridimensionales de esta enzima resueltas; en concreto s\u00f3lo dos, las de los organismos escherichia coli y bacillus subtilis, pero ninguno de ellos presenta comportamiento extrem\u00f3filo. A pesar de ello, las isocitrato deshidrogenasas dim\u00e9ricas presentan un elevado grado de identidad de secuencia, lo que permite trabajar con estas dos enzimas como modelos.       En particular, en la enzima de e. Coli, se han realizado estudios de regulaci\u00f3n por fosforilaci\u00f3n, de mutag\u00e9nesis para producir el cambio en la especificidad del coenzima a nad y para identificar los amino\u00e1cidos implicados en la uni\u00f3n del sustrato, as\u00ed como ensayos cristalogr\u00e1ficos con la consecuente resoluci\u00f3n de la estructura tridimensional.    En este trabajo, se han realizado estudios filogen\u00e9ticos, bioqu\u00edmicos y f\u00edsico-qu\u00edmicos como fluorescencia, dicro\u00edsmo circular y ultracentrifugaci\u00f3n diferencial, as\u00ed como ensayos de mutag\u00e9nesis dirigida para producir el cambio de especificidad de la enzima por el coenzima a nad; adem\u00e1s, se ha realizado el modelado te\u00f3rico de la enzima, tanto en su forma nativa como mutante. Para realizar la gran mayor\u00eda de estos estudios es necesario obtener gran cantidad de isocitrato deshidrogenasa que, por cultivo del organismo de procedencia, hfx. Volcanii, ser\u00eda muy complicado de conseguir y requerir\u00eda mucho tiempo y esfuerzo. Para ello se ha clonado y expresado la enzima de forma heter\u00f3loga en e.
