La muerte por apoptosis est\u00e1 en el origen y mecanismo fisiopatol\u00f3gico de diversas enfermedades hep\u00e1ticas en las que el tratamiento con adomet ejerce efectos beneficiosos. En el presente trabajo se ha analizado el efecto de aclomet en la apoptosis inducida por el \u00e1cido okadaico (ao) en hepatocitos primarios de rata en cultivo, y en lineas celulares de hepatoma humano. Adomet protege a los hepatocitos de la apoptosis inducida por el ao de manera dependiente de la dosis. Asimismo, atenua la liberaci\u00f3n de citocromo c de la mitocondria, la activaci\u00f3n de la caspasa 3, y la fragmentaci\u00f3n de la poli(adp-ribosa) polimerasa. Estos efectos son independientes de la s\u00edntesis de glutation y de las reacciones de metilaci\u00f3n, siendo mimetizados por la 5-metiltioadenosina (mta), un metabolito derivado de adomet. Es la propia mta, y no metabolitos derivados de este compuesto, la responsable del efecto antiapopt\u00f3tico en los hepatocitos primarios. Por otra parte, ni adomet ni mta protegen de la apoptosis inducida por el ao en las c\u00e9lulas de hepatoma humano huh7 y hepg2, presentando ambos compuestos por s\u00ed mismos, un efecto proapopt\u00f3tico en ambos tipos celulares. As\u00ed pues, adomet ejerce efectos opuestos en hepatocitos primarios y en c\u00e9lulas de hepatoma, que pueden estar mediados por su transformaci\u00f3n en mta. El efecto proapopt\u00f3tico del ao es debido a la activaci\u00f3n de la v\u00eda jnk\/ap-l. El pretratamiento de los hepatocitos con adomet o mta, previene la fosforilaci\u00f3n de la 3hk y de c-jun, as\u00ed como la actividad de uni\u00f3n del factor ap-1. Sin embargo, ni adomet ni mta previenen la fosforilaci\u00f3n de la jnk y de c-jun, ni la activaci\u00f3n del factor ap-1 inducida por el ao en la l\u00ednea celular de hepatoma huh7.
