El vca y el phi se miden simult\u00e1neamente, en c\u00e9lulas aisladas, a partir de los cambios en la fluorescencia del indicador bcecf, tras la incorporaci\u00f3n de \u00e9ste a las c\u00e9lulas. Para los registros se utiliz\u00f3 una t\u00e9cnica de microscop\u00eda de fluorescencia cuanti-tativa desarrollada y validada en nuestro laboratorio. Se demuestra que la exposici\u00f3n transitoria de las c\u00e9lulas a soluciones isosm\u00f3ticas conteniendo nh3\/nh4+ (nh4cl: 0,5-30 mm) produce, adem\u00e1s de los cambios esperados en el phi, cambios en el vca, en los que se distinguen 4 fases: 1) un aumento r\u00e1pido; 2) una disminuci\u00f3n reguladora del volumen (drvi); y tras la eliminaci\u00f3n del pulso de nh4cl: 3) un encogimiento r\u00e1pido, seguido de 4) un aumento regulador del volumen (arvi). La ocurrencia de cambios en el volumen celular ante la exposici\u00f3n a sales de amonio, aunque predicha desde hace m\u00e1s de 80 a\u00f1os (h. M. Jacobs, 1940), no hab\u00eda sido demostrada hasta el presente. A partir de la caracterizaci\u00f3n de estos cambios, se investigan los mecanismos subyacentes a los mismos. se concluye que una parte importante del aumento inicial del vca se de-be a la acumulaci\u00f3n intracelular de nh4+ y un hipot\u00e9tico ani\u00f3n acompa\u00f1ante, a\u00fan no identificado, que ser\u00eda necesario para preservar la electroneutralidad macrosc\u00f3pica del interior celular. El modelo presentado tiene su correlato fisiopatol\u00f3gico en el edema cerebral asociado con las hiperamonemias agudas en el hombre, y constituye una herramienta \u00fatil para el estudio de los
