Durante la transformaci\u00f3n neopl\u00e1stica,  las c\u00e9lulas adquieren mutaciones gen\u00e9ticas que les permiten escapar de los mecanismos de control de la proliferaci\u00f3n celular. C\u00e9lulas primaria murinas pueden ser transformadas en tumorales mediante la coexpresi\u00f3n de dos oncogenes cooperantes como c-myc y h-rasv12. Sin embargo, experimentos similares en c\u00e9lulas humanas no han tenido \u00e9xito en la obtenci\u00f3n de c\u00e9lulas transformadas. Estas diferencias pueden atribuirse, al menos en parte, a las diferencias que existen entre c\u00e9lulas humanas y de rat\u00f3n en cuanto a su biolog\u00eda telom\u00e9rica. Mientras que c\u00e9lulas murinas expresan telomerasa y sus tel\u00f3meros son extremadamente largos, las c\u00e9lulas humanas carecen de telomerasa y sus tel\u00f3meros se acortan tras cada ciclo de divisi\u00f3n celular. Este acortamiento telom\u00e9rico supone una barrera que limita la capacidad proliferativa de las c\u00e9lulas humanas. la importancia de la telomerasa en el proceso de conversi\u00f3n de una c\u00e9lula humana  normal en tumoral se pone de manifiesto en que la mayor\u00eda de los tumores humanos presentan elevados niveles de actividad telomerasa.  Los modelos existentes de obtenci\u00f3n de c\u00e9lulas humanas mediante la alteraci\u00f3n de elementos gen\u00e9ticos definidos inclu\u00edan la activaci\u00f3n de un mecanismo de mantenimiento telom\u00e9rico como un requisito necesario para la transformaci\u00f3n celular. En nuestro trabajo hemos descrito un modelo alternativo de transformaci\u00f3n celular que  no requiere la activaci\u00f3n de telomerasa o de otro mecanismo alternativo de mantenimiento telom\u00e9rico.
