En el plegamiento del d\u00edmero de alfa- y beta-tubulina se han identificado diferentes cofactores implicados en el plegamiento y funci\u00f3n de la alfa- y beta-tubulina, permitiendo comprender la integraci\u00f3n de las dos rutas de plegamiento de tubulina, las cuales convergen de tal manera que el plegamiento de una requiere la presencia de diferentes cofactores y de la otra subunidad. el cofactor c funciona como una gap (gtp-asa activating protein) para la beta-tubulina dirigiendo la liberaci\u00f3n del heterod\u00edmero del complejo alfa\/beta\/d. la primera parte de esta  tesis se ha centrado en el estudio del cofactor c y del complejo p14\/beta tubulina).  se han obtenido y purificado por afinidad anticuerpos espec\u00edficos contra el cofactor c humano completo y contra el dominio carboxi-terminal del cofactor c murino. La expresi\u00f3n del cofactor c mediante la infecci\u00f3n con baculovirus recombianantes da lugar a dos formas moleculares capaces ambas de activar la liberaci\u00f3n del d\u00edmero de tubulina del supercomplejo c300. la diferencia entre ambas formas no es debido a una farnesilaci\u00f3n, como sugieren los experimentos de maldi-tof ni a una fosforilaci\u00f3n. La expresi\u00f3n del cofactor c tanto a nivel de rna como de prote\u00edna se localiza principalmente en el test\u00edculo y est\u00e1 adem\u00e1s temporalmente correlacionada su expresi\u00f3n con la de los isotipos de tubulina b3, a3\/7 y cofactor a, coincidiendo su mayor aumento a nivel de expresi\u00f3n con el inicio de la expermiog\u00e9nesis. la sobreexpresi\u00f3n del cofactor c en las c\u00e9lulas hela no produce ninguna alteraci\u00f3n fenot\u00edpica apreciable, por lo que no parece que el cofactor c, por s\u00ed solo, sea una prote\u00edna reguladora de los procesos de cat\u00e1strofe de microt\u00fabulos. Estudios en cultivos primarios muestran una expresi\u00f3n mayoritaria del cofactor c en neuronas.
