Coli silvestres da lugar a anticuerpos no funcionales. Esto se debe a la imposibilidad de que se formen, en el ambiente reductor del citoplasma, los enlaces disulfuro que contienen los anticuerpos. La formaci\u00f3n de estos enlaces es necesaria para el plegamiento y la actividad de la mayor\u00eda de los dominios inmunoglobulina (ig). En este trabajo se investiga la formaci\u00f3n de puentes disulfuro y la funcionalidad de los anticuerpos monocadena (scfv), en el citoplasma de estirpes de e. Coli con mutaciones en genes que forman parte los dos principales sistemas intracelulares de reducci\u00f3n de enlaces disulfuro (la ruta de las tiorredoxinas, y la de las glutarredoxinas). Se ha examinado asimismo el efecto de la coexpresi\u00f3n de diferentes chaperonas de puentes disulfuro, en trans o fusionadas al scfv. Los resultados de esta tesis demuestran que la forma m\u00e1s eficiente de producir intracuerpos funcionales, con puentes disulfuro en sus dominios ig, tiene lugar en cepas mutantes en los genes que codifican a la tiorredoxina reductasa (trxb) y la glutation oxidorreductasa (gor) en las cuales el scfv se produce fusionado al extremo c-terminal de la tiorredoxina 1 (trx1).  hemos transformado una cepa trxb gor con genotecas de fragmentos de dna, que codificin a diferentes colecciones de scfvs, con la intenci\u00f3n de buscar anticuerpos capaces de inhibir o activar un proceso de transcripci\u00f3n desde un promotor concreto. En estas b\u00fasquedas hemos encontrado varios anticuerpos capaces de activar o de inhibir el mecanismo de transcripci\u00f3n desde un promotor dependiente del factor<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos
