an\u00e1logos"},"content":{"rendered":"<h2>Tesis doctoral de <strong> Xenxo P\u00e9rez Garc\u00eda <\/strong><\/h2>\n<p>La mayor\u00eda de las funciones biol\u00f3gicas que desempe\u00f1a la vitamina d3 tienen lugar a trav\u00e9s de su forma hormonal activa , la 1a,25-dihidroxivitamina-d3.Entre estas funciones biol\u00f3gicas se encuentran la homeostasis del calcio y del f\u00f3sforo , pero recientemente se ha descubierto que la vitamina d3 interviene en otros procesos como la diferenciaci\u00f3n y maduraci\u00f3n celular, procesos antiproliferativos, regulaci\u00f3n del sistema inmunol\u00f3gico , etc.  el primer objetivo de esta tesis es la s\u00edntesis de una serie de an\u00e1logos de la 1a,25-(oh)2-d3 de cadena lateral r\u00edgida o semirr\u00edgida con el hidroxilo orientado hacia la regi\u00f3n que ocupa en el cristal de moras con el doble prop\u00f3sito de : 1)saber si la regi\u00f3n definida en el cristal es biol\u00f3gicamente activa y 2)obtener an\u00e1logos con propiedades selectivas y , en consecuencia, de potencial valor terp\u00e9utico (c\u00e1ncer,psoriasis,osteoporosis,etc).Para la s\u00edntesis de estos an\u00e1logos se ha utilizado : a)un acoplamiento tipo wittig-horner para generar el sistema tri\u00e9nico de la vitamina d;b)en el caso de los an\u00e1logos r\u00edgidos, un acoplamiento catalizado por metales (pd,cu)para la s\u00edntesis de la cadena lateral \u00f3 en el caso delos an\u00e1logos semirr\u00edgidos, una reacci\u00f3n ald\u00f3lica para generar el anillo adicional.Los ensayos biol\u00f3gicos realizados a los an\u00e1logos de cadena lateral r\u00edgida demostraron que son capaces de inducir la transcripci\u00f3n gen\u00e9tica en c\u00e9lulas procedentes de c\u00e1ncer de colon en un orden similar o superior que la propia hormona calcitriol presentando efectos calc\u00e9micos de cadena muy inferiores a los que produce la 1a,25-(oh)2-d3-las pruebas biol\u00f3gicas realizadas a los an\u00e1logos de cadena lateral semirr\u00edgida indican que  estos compuestos tienen una baja afinidad de enclace al vdr y a la dbp mucho menor que el calcitriol.Sin embargo , presentan un efecto antiproliferativo en
