Dolores <\/strong><\/h2>\n<p>Los tumores astrociticos constituyen, a pesar de los avances de las1\u00e9cnicas quir\u00fargicas, un problema sin resolver. Las formas de mayor grado de malignidad siguen sin tener una supervivencia mayor de 9-12 meses. El conocimiento molecular de las 21teraciones que aparecen en dichos tumores de forma secuencial es b\u00e1sico para iniciar un tratamiento quimio o radioter\u00e1pico, dado que se han encontrado distintos grados de respuesta seg\u00fan dichas alteraciones.  en nuestra serie hemos incluido 86 tumores astroc\u00edticos de los distint0s grados de malignidad, procediendo a buscar en ellos perdidas de heterocigosidad, hipermetilaci\u00f3n y mutaciones, as\u00ed como amplificaci\u00f3n y sobreexpresi\u00f3n g\u00e9n\u00edca, en genes y regiones involucradas en la transformaci\u00f3n maligna de este tipo tumoral.  las conclusiones de nuestro estudio son las siguientes:  1. Las p\u00e9rdidas de heterocigosidad de los brazos cromos\u00f3micos 9p, 1op y 10q-se relacionan significativamente con un alto grado de malignidad de los tumores astroc\u00edticos, especialmente en la v\u00eda de desarrollo de los glioblastomas \u00abde novo\u00bb. Las perdidas en 19q, en cambio, estar\u00edan relacionadas con la v\u00eda progresiva de evoluci\u00f3n tumora!. Menos significativas parecen ser las p\u00e9rdidas en 1p en el desarrollo de tumores astrociticos.  2. Las mutaciones en pten, as\u00ed como la amplificaci\u00f3n del gen egfr son sucesos relacionados con la loh del cromosoma 10, apareciendo posteriormente a dichas deleciones vinculadas a un aumento en el grado de malignidad tumoral.  3. Las mutaciones de tp53, la amplificaci\u00f3n de pdgfr-a y de los genes localizados en la regi\u00f3n 12q 13 se encuentran implicados en la aparici\u00f3n de las formas tumorales astroc\u00edticas de grado 11.  4. El gen mdm2 parece ser la diana del proceso de amplificaci\u00f3n, en estadios tempranos, de la regi\u00f3n 12q13 en un subgrupo de tumores que no presentan amplificaci\u00f3n de mdm4 ni mutaciones en tp53.
