En la -carcinog\u00e9nesis, tgf-beta desempe\u00f1a un doble papel, actuando como un supresor tumoral en los estadios m\u00e1s tempranos, pero estimulando la progresi\u00f3n maligna en los estadios m\u00e1s avanzados. Tgf-beta activa un complejo mecanismo de transducci\u00f3n de se\u00f1ales. La principal v\u00eda de respuesta al factor est\u00e1 mediada por las proteinas-de la familia smad. sin embargo, otras rutas, como mapk, las quinasas dependientes de estr\u00e9s, rho gtpasas o pi3k, pueden ser activadas por tgf-beta en distintos tipos celulares. En este trabajo se muestra que tgf-b~ta1 activa la se\u00f1alizaci\u00f3n ras\/mapk en queratinocitos, independientemente de.La v\u00eda smad. Mediante la transfecci\u00f3n de una construcci\u00f3n dominante-negativa de smad4, o de smad4 silvestre, en c\u00e9lulas de distintos tipos de carcinoma murino y humano mostramos, adem\u00e1s, que existen interrelaciones muy complejas de smad4 con otras vias de se\u00f1alizaci\u00f3n. As\u00ed, en los transfectantes de queratinocitos transformados pdv que ten\u00edan bloqueado funcionalmente smad4 se produjo una hiperactivaci\u00f3n de ras\/mapk y una aceleraci\u00f3n de la progresi\u00f3n maligna; mientras que la p\u00e9rdida funcional de smad4 en c\u00e9lulas de carcinoma pancre\u00e1tico panc-1 condujo a una. Aparente reducci\u00f3n de la tumorogenicidad asociada con una disminuci\u00f3n de los niveles y de la se\u00f1alizaci\u00f3n nuclear de beta-catenina, probalmente debida a un incremento de su degradaci\u00f3n por el proteasoma. en c\u00e9lulas de carcinoma de colon sw480 (que son deficientes para la expresi\u00f3n de smad4)la reexpresi\u00f3n de smad4 disminuy\u00f3 los niveles de ras activo y condujo a una reversi\u00f3n parcial de la malignidad. Estos resultados sugieren la existencia de un nuevo tipo de interrelaci\u00f3n entre smad4 y ras, en c\u00e9lulas de carcinoma epitelioide y de carcinoma de colon que contienen un oncog\u00e9n ras,
