{"id":73100,"date":"2005-09-03T00:00:00","date_gmt":"2005-09-03T00:00:00","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/identificacion-de-quimiosensibilizadores-de-fenotipo-mdr-a-traves-de-quimiotecas-sinteticas-y-naturales\/"},"modified":"2005-09-03T00:00:00","modified_gmt":"2005-09-03T00:00:00","slug":"identificacion-de-quimiosensibilizadores-de-fenotipo-mdr-a-traves-de-quimiotecas-sinteticas-y-naturales","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/ciencias-medicas\/identificacion-de-quimiosensibilizadores-de-fenotipo-mdr-a-traves-de-quimiotecas-sinteticas-y-naturales\/","title":{"rendered":"Identificaci\u00f3n de quimiosensibilizadores de fenotipo mdr a trav\u00e9s de quimiotecas sinteticas y naturales."},"content":{"rendered":"<h2>Tesis doctoral de <strong> Mar\u00eda  Jose Abad Merin <\/strong><\/h2>\n<p>Para identificar compuestos quimiosensibilizadores destinados a paliar la aparici\u00f3n del fenotipo de resistencia a m\u00faltiples f\u00e1rmacos (mdr) asociado a la resistencia tumoral a los f\u00e1rmacos antineopl\u00e1sticos cl\u00e1sicos, se realizaron las siguientes aportaciones:  &#8211; se estableci\u00f3 primeramente un ensayo de cribado celular de alto rendimiento mediante citometr\u00eda de flujo basado en la acumulaci\u00f3n intracelular de la antraciclina daunomicina.  &#8211; se utiliz\u00f3 dicho ensayo para cribar y refinar posteriormente mediante qu\u00edmica combinatoria dos quimiotecas sint\u00e9ticas de n-alquilglicinas (5120 compuestos) y 2,5-dicetopiperacinas (165 compuestos), as\u00ed como diversas fracciones de un extracto vegetal obtenidas por hplc. El modelo celular de ensayo consisti\u00f3 en dos l\u00edneas hematopoy\u00e9ticas de rat\u00f3n (l1210)y humana(hl-60) disponibilidad a partir de ambas l\u00edneas de sublineas resistentes a los f\u00e1rmacos antineopl\u00e1sicos cl\u00e1sicos y con fenotipo mdr, obtenidas mediante presi\u00f3n selectiva (l1210r y hl60-r).  &#8211; de la quimioteca de n-alquilglicinas se han obtenido dos quimiosensibilizadores activos. La quimioteca de 2,5- dicetopiperacinas requiere un posterior refinamiento.
