La eficacia de este mecanismo reside en su capacidad para activarse espec\u00edficamente sobre los agentes pat\u00f3genos, limitando el da\u00f1o asociado a la activaci\u00f3n incontrolada del complemento sobre los tejidos propios. Dicha especificidad se basa en la existencia de una serie de prote\u00ednas reguladoras, muchas de las cuales est\u00e1n codificadas por genes localizados. En la regi\u00f3n gen\u00f3mica iq32, dentro del agrupamiento g\u00e9nico rca (regulators ofcomplement activation).  en la presente tesis doctoral he aplicado el m\u00e9todo de descomposici\u00f3n de la varianza.Para el an\u00e1lisis de los factores gen\u00e9ticos responsables de la variabilidad en los niveles plasm\u00e1ticos de dos reguladores del complemento del rca (c4bp y factor h). Adem\u00e1s se evaluaron las posibles implicaciones patol\u00f3gicas asociadas a la variabilidad en estos fenotipos.  la prote\u00edna c4b-binding protein (c4bp) tiene la capacidad de interferir en el sistema de la coagulaci\u00f3n a trav\u00e9s de su interacci\u00f3n con la prote\u00edna s (ps). En este trabajo se ha aplicado el modelo umbral o threshold con el fin de analizar la contribuci\u00f3n de los fenotipos plasm\u00e1ticos de c4bp y ps a la susceptibilidad a padecer trombosis. Adem\u00e1s se ha estudiado el papel de las proteinas reguladoras del complemento del rca en la predisposici\u00f3n al s\u00edndrome hem\u00f3l\u00edtico ur\u00e9mico at\u00edpico (ahus), una enfermedad ocasionada por la activaci\u00f3n incontrolada del complemento sobre el endotelio renal como consecuencia de alteraciones funcionales en proteinas del rca. En este contexto nuestro  estudio se ha realizado considerando las variaciones .Cuantitativas (niveles de expresi\u00f3n) y cualitativas (d\u00e9ficit funcional) gen\u00e9ticamente determinadas en
