Los est\u00edmulos apopt\u00f3sicos inducen , la traslocaci\u00f3n de miembros pro-apopt\u00f3sicos de la familia de bcl-2 a la membrana mitocondrial externa donde fonnan canales i\u00f3nicos que pueden contribuir a disipar el potencial transmembrana de la mitocondria y favorecer la liberaci\u00f3n de cito cromo c. En el citosol, esta proteina se une al factor activador de apoptosis (apaf-l) para folmar un complejo denominado apoptosoma, el cual activa una familia de proteasa\u00c2\u00a7 denominada caspasas. Finalmente las caspasas hidrolizan una serie de prote\u00ednas clave para la supervivencia celular y la c\u00e9lula muere de folma organizada y nonecr\u00f3tica. actualmente la implicaci\u00f3n de la apoptosis en c\u00e1ncer y enfermedades neurodegenerativas est\u00e1 bien establecida.  resultados  la identi\u00f1caci\u00f3n de agentes que puedan modular el proceso de apoptosis es de gran inter\u00e9s farrnacol\u00f3gico. En primer lugar, se ha puesto a punto un ensayo bioquimico de reconstituci\u00f3n de apoptosomas purificados y seguimiento de la activaci\u00f3n de caspasas a partir de sus componentes b\u00e1sicos expresados in vitro. Pare ello se ha puesto a punto la expresi\u00f3n de la proteina (apaf-1 )en el sistema de expresi\u00f3n con bacu1ovirus y se ha caracterizado y optimizado la cin\u00e9tica de la expresi\u00f3n para la obtenci\u00f3n de prote\u00edna funcional. Para identi\u00f1car mol\u00e9culas que modulen la folmaci\u00f3n del apoptosoma hemos utilizado como estrategia la q1ri1i\u00faca combinatoria.
