La v\u00eda de formaci\u00f3n del \u00e1cido retinoico incluye dos pasos de oxidaci\u00f3n, de retinol y retinal, y de retinal a \u00e1cido retinoico. Se han descrito varias enzimas que pueden participar en el primer paso, c\u00f3mo las adh y las rdh, pero se desconoce la contribuci\u00f3n de cada enzima en el proceso.  se ha clonado, expresado y purificado adh1b1, adh2b2 y adh4 humanas y, tambi\u00e9n adh1 y adh4 de rat\u00f3n, que por primera vez, se han caracterizado en su forma recombinante. Se han determinado las constantes cin\u00e9ticas de todas estas adhs con todo-trans- y 7-cis-, 9-cis-, 11-cis- y 13-cis-retinol y los correspondientes retinales. En general, las adh4 humanas y de rat\u00f3n muestran constantes cin\u00e9ticas similares, y son m\u00e1s eficientes que las adh1.  todas las adhs utilizan como sustratros tanto 11-cis-retinol como 11-cis-retinal, compuestos relevantes en el ciclo visual. Se ha observado que la adh4 muestra especificidad para la oxidaci\u00f3n de 11-cis-retinol sobre la reducci\u00f3n de 11-cis-retinal, una propiedad \u00fanica entre todas las adhs para cualquier pareja de sustratos alcohol\/aldeh\u00eddo.  mediante m\u00e9todos de simulaci\u00f3n molecular, y colonaci\u00f3n, expresi\u00f3n y caracterizaci\u00f3n del mutante de la adh4 humana m141l, hemos demostrado que el residuo 141, situado en la regi\u00f3n mediana del t\u00fanel hidrof\u00f3bico del sitio activo de la adh, es esencial para definir esta especificidad de la adh4 sobre las adh1. La inmunolocalizaci\u00f3n de la adh4 en el epitelio pigmentado y en muhcas capas de la retina apoya la participaci\u00f3n de la adh4 en diferentes reacciones con retinoides. La actividad citos\u00f3lica de la adh4 en el epitelio pigmentado puede ser complementaria a la actividad 11-cis-retinol deshidrogenasas de la rdh5, necesaria para completar el ciclo visual, y puede
