Para ello , se han clonado y caracterizado los genes hppd1 y tmt1 de c. Reinhardtii, genes que codifican la p-hidroxifenilpiruvato dioxigenasa (hppd) y la  gamma-tocoferol metiltransferasa (gamma-tmt), primera y \u00faltima enzima de la ruta de s\u00edntesis de alfa-tocoferol. La clanaci\u00f3n de hppd1 en un vector de expresi\u00f3n ha permitido purificar dicha enzima en forma de prote\u00edna del fusi\u00f3n y su caracterizaci\u00f3n d\u00edsico-qu\u00edmica y cin\u00e9tica. la enzima tiene una masa molecular de 47 kda y en condiciones nativas, se organiza en tetr\u00e1meros de subunidades del mismo tama\u00f1o no uniddas entre s\u00ed por puentes disulfuro.Se ha estudiado el papel que la temperatura y la luz tienen sobre la regulaci\u00f3n de la acumulaci\u00f3n y s\u00edntesis de alfa, beta, y gamma-tocoferon en c.Reinhardtii. La determinaci\u00f3n mediante hplc de la acumulaci\u00f3n de los distintos tocoferoles y el an\u00e1lisis mediante northern-blot de los genes hppd1 ytmt1 revelan que a baja temperatura se acumula espec\u00edficamente gamma-tocoferol, mientras que a temperaturas superiores se acumulan alfa-tocoferol. ninguno de os tocoferoles vari\u00f3 significativamente tras exponer las c\u00e9lulas a una irradiancia mayor ni tras someterlas a oscuridad; sin embargo dichas condiciones , s\u00ed que se afect\u00f3 la expresi\u00f3n de happd1 y tmt1. La respuesta diferencial en la acumulaci\u00f3n de alfa y gamma-tocoferol ante la variaci\u00f3n de dichos par\u00e1metros ambientales , unido a las diferentes propiedades estructurales y f\u00edsico-qu\u00edmicas , sugiere que alfa y gamma-tocoferol realizan funciones diferentes, bien debido a una localizaci\u00f3n diferente o a que participan en procesos distintos. Finalmente , se han transformado c\u00e9lulas de c.Reinhardtii con una construcci\u00f3n que contine el gen hppd1 bajo el cotrol de promotor 35s.Los resultados
