Esta es una enfermedad hereditaria caracterizada por una degeneraci\u00f3n selectiva de las neuronas gaba\u00e9rgicas del n\u00facleo estriado. Para poder desarrollar terapias de reemplazo celular basadas en los transplantes de c\u00e9lulas madre es necesario controlar el proceso de diferenciaci\u00f3n de las c\u00e9lulas madre hacia el fenotipo neuronal gaba\u00e9rgico.  en la presente tesis hemos estudiado los mecanismos de supervivencia de las c\u00e9lulas madre en cultivo y los factores implicados en la diferenciaci\u00f3n selectiva hacia el fenotipo gaba\u00e9rgico. Hemos desarrollado un protocolo de diferenciaci\u00f3n in vitro basado en la adici\u00f3n secuencial de \u00e1cido retinoico (ra) y cloruro pot\u00e1sico (kci). El ra promueve la supervivencia celular e induce, un fenotipo neuronal. El tratamiento con kci induce selectivamente el fenotipo gaba\u00e9rgico. Mediante este procedimiento se generan un elevado n\u00famero de neuronas gaba\u00e9rgicas maduras, post-mit\u00f3ticas y funcionales, de forma eficaz y homog\u00e9nea, a partir de una l\u00ednea de c\u00e9lulas madre en cultivo. el trasplante de estas c\u00e9lulas pre-diferenciadas in vitro en el cerebro adulto de rata y en un modelo excitot\u00f3xico de la enfermedad de huntington demuestran un buen grado de diferenciaci\u00f3n, supervivencia e integraci\u00f3n funcional. Las c\u00e9lulas muestran una supervivencia a largo plazo, conservan el fenotipo neuronal gab\u00e9rgico y elaboran proceso neur\u00edticos asociados a posibles contactos pre- y post-sin\u00e1pticos.  estos resultados apoyan la posibilidad de desarrollar terapias celulares basadas en la diferenciaci\u00f3n de c\u00e9lulas madre para el tratamiento de enfermedades
