Se determin\u00f3 que el n\u00famero de neuronas de la capa molecular se encontraba en torno a las 14000-17000 neuronas sin apreciarse diferencias estad\u00edsticamente significativas entre animales perinatales, j\u00f3venes y adultos.  la primera clasificaci\u00f3n de las neuronas de capa molecular se realiz\u00f3 empleando t\u00e9cnicas fisiol\u00f3gicas cl\u00e1sicas (t\u00e9cnica histoqu\u00edmica de la nadph-diaforasa, tinci\u00f3n de golgi, inmunotinciones frente a prote\u00ednas quelantes de calcio y tinciones de nissl). La observaci\u00f3n de las prepaciones demostr\u00f3 la existencia de ocho arquetipos denominados, en funci\u00f3n de su morfolog\u00eda, como: verticales, globosas, sarmentosas, granulares ect\u00f3picas, piramidales invertidas, polimorfas, horizontales peque\u00f1as y horizontales grandes. Cada tipo neuronal present\u00f3 una ubicaci\u00f3n preferencial en alguna de las subcapas de capa molecular: en capa molecular externa los tipos m\u00e1s frecuentes son las neuronas horizontales peque\u00f1as, sarmentosas y piramidales invertidas; en capa molecular intermedia las neuronas polimorfas, verticales y globosas; y en capa molecular interna las granulares ect\u00f3picas y horizontales grandes. Todos los tipos mantienen poblaciones constantes durante la vida del animal excepto las granulares ect\u00f3picas que aumentan ligeramente y las horizontales peque\u00f1as que disminuyen hasta la tercera semana de vida del animal. La ubicaci\u00f3n debe estar relacionada con la regulaci\u00f3n de la aferencia propia de la subcapa que ocupan. Las neuronas globosas y granulares ect\u00f3picas expresan calbindina, las polimorfas horizontales grandes y algunas horizontales peque\u00f1as parvalb\u00famina, y las horizontales peque\u00f1as y verticales calretinina.  la clasificaci\u00f3n fue confirmada mediante la t\u00e9cnica de inyecci\u00f3n intracelular en tejido fijado y vivo.
