Los eventos que ocurren durante la hipoxia y la subsiguiente repercusi\u00f3n, producen inflamaci\u00f3n, necrosis tubular, disfunci\u00f3n renal, fibrosis reversible y posible progresi\u00f3n a fracaso renal cr\u00f3nico. entre los mediadores implicados en este da\u00f1o est\u00e1n la angiotensina ii, la citocina tgfb1 y las kininas, todos ellos interrelacionados entre s\u00ed y con la hipoxia. M\u00e9todos: se produce isquemia por oclusi\u00f3n de un ped\u00edculo renal durante 45min en ratas o 25min en ratones. Para confirmar los resultados m\u00e1s relevantes se emplea otro modelo de fracaso renal agudo nefrot\u00f3xico por cloruro de mercurio (inyecci\u00f3n subcut\u00e1nea; dosis 2 u 8 mg\/kg para ratas o ratones, respectivamente). Se determina la funci\u00f3n y estructura renal (0,5-14 d\u00edas), y la expresi\u00f3n g\u00e9nica de marcadores de fibrosis y de inflamaci\u00f3n (2-3 d\u00edas). Objetivos: se estudia el papel de la endoglina, una prote\u00edna accesoria receptor para tgfb1, del sistema kinina-kalicre\u00edna y del receptor b1 de bradikinina en el fracaso renal agudo por i-r.  Se emplean animales normales y modificados gen\u00e9ticamente: rat\u00f3n haploinsuficiente para endoglina (eng+\/-), rata transg\u00e9nica de kalikre\u00edna tisular humana (tgr tklk-1) y rat\u00f3n carente del gen para el receptor b1 de bradikinina (b1-\/-). Resultados y discusi\u00f3n: la endoglina aumenta en los ri\u00f1ones isqu\u00e9micos de animales normales sometidos a i-r. La falta de endoglina en los ratones eng+\/- protege del da\u00f1o por isquemia; as\u00ed, la funci\u00f3n y estructura renal son normales tras i-r, y no aumentan los marcadores de fibrosis o de inflamaci\u00f3n comparado con los ratones normales. La hiperexpresi\u00f3n de la kalikre\u00edna renal empeora el da\u00f1o estructural y funcional tras i-r renal, y aumentan los marcadores de fibrosis e inflamaci\u00f3n. Todos estos resultados se confirman en el modelo de nefrotoxicidad por mercurio.
