Este proceso, conocido como \u00abshedding\u00bb (del ingl\u00e9s desporjarse o deshacerse), remodelada la superficie de las c\u00e9lulas y regula la funci\u00f3n de un n\u00famero diverso de prote\u00edna de transmembrana que incluyen  factores de crecimiento, citoquinas, mol\u00e9culas de adhesi\u00f3n, receptores de factores de crecimiento y de citoquinas. Durante la d\u00e9cada de los ochenta y noventa se puso de manifiesto el papel  que jugaban metaloproteasas dependientes de zinc en la realizaci\u00f3n de la mayor\u00eda, si no es de todos, los procesos de shedding descritos, ya que inhibidores de metaloproetasas bloquean eficientemente este proceso. No obstante, las metaloproteasas implicadas en el shedding de ectodominios no se revelaron hasta que en 1997, se descubri\u00f3 el miembro n\u00famero 17 de la familia adma y se identific\u00f3 como la metaloproteasas responsable del shedding de tnf-alfa.  las c\u00e9lulas derivadas de ratones que tienen una inactivaci\u00f3n gen\u00e9tica de esta metaloproteasa (ratones tace zn\/zn) muestran inhibida la producci\u00f3n de tnf-alfa soluble. Esta mealoproteasas se denomin\u00f3 adma 17 o tace (tumor necrosis factor-alfa converting enzyme). El fenotipo de los ratones tace zn\/zn muestra que tace es esencial en el desarrollo de estos animales y el uso de fibroblastos derivados de ellos demuestra que es responsable del shedding de un gran n\u00famero de prote\u00ednas de transmembrana de funci\u00f3n y estructuras muy distintas. Dada esta variedad de sustratos, entre ellos ligandos de la familia del egf, es razonable conjeturar que la actividad de tace estar\u00e1 bajo un riguroso control para prevenir la prote\u00f3lisis no deseada y la activaci\u00f3n de determinados receptores.
