En este mecanismo de entrada parecen estar implicados enzimas hidroliticos que degradan la pared celular. El objetivo de este trabajo ha sido determinar la existencia de enzimas pecticos en leguminosas y analizar su posible implicacion en la simbiosis. Se ha clonado el gen de poligalacturonasa (pg) de tomate bajo el control de los promotores tac y trc y expresado en e. Coli y en rhizobium, lo que ha permitido poner a punto las tecnicas de deteccion de actividad pg. Se han aislado y secuenciado tres productos de pcr, con homolog\u00eda a regiones conservadas entre diferentes pgs de plantas, a partir de adn genomico de m. Truncatula (e2 y e3) y de adnc de polen de m. Sativa (pl1) y otros dos a partir de trifolium subterraneum (tf6 y tf7). E2 y pl1 pertenecen a un gen de pg que forma parte de una peque\u00f1a familia genica y que se expresa fuertemente en flores maduras y no en raices o nodulos. E3 pertenece a un gen de pg que se encuentra en una sola copia y se expresa en raices inoculadas con la cepa silvestre de r. Meliloti y muy debilmente en raices no inoculadas o inoculadas con un mutante nod-. La expresion se detecta en raices un dia despues de la inoculacion. Los transcritos no se detectan en flores y si en nodulos inducidos por la cepa silvestre. Se ha detectado una induccion en nodulos de 31 dias inducidos por la cepa silvestre con respecto a nodulos de 7 dias y, ademas, una mayor expresion en nodulos inducidos por un mutante fix exo- con respecto a los nodulos inducidos por la cepa silvestre. Tambien se ha detectado una ligera induccion de la expresion en raices heridas e inoculadas con pseudomonas 17 horas despues de haberse tratado. Se ha clonado y secuenciado parcialmente el gen de pg de m. Sativa de expresion en nodulo y raiz.
